【摘要】：目的探討正常人血小板中是否存在Aβ蛋白淀粉樣變成分。方法篩選正常人群340例,采用Sysmex-200型血細(xì)胞分析儀測(cè)定血液血小板數(shù)量(PLT)、血小板分布寬度(PDW)、平均血小板體積(MPV)、血小板比容(PCT)和大型血小板比例(P-LCRs),進(jìn)一步提純血小板,將血小板分成兩部分,一部分用于提取血小板膜蛋白,另一部分用硫磺素T(Th T)熒光探針檢測(cè)血小板膜上淀粉樣變�？笰β抗體檢測(cè)淀粉樣變前多肽的免疫反應(yīng)。Western印跡觀察Aβ蛋白分子聚合。結(jié)果 Th T熒光探針檢測(cè)血小板幾乎全部顯示陽(yáng)性反應(yīng)。進(jìn)一步抗Aβ免疫組織化學(xué)實(shí)驗(yàn)發(fā)現(xiàn)綠色熒光與Aβ抗體反應(yīng)陽(yáng)性部位重疊。SDS-PAGE分析血小板提取的蛋白成分含有大分子Aβ聚合物。結(jié)論正常人血小板中存在Aβ淀粉樣變成分,可能參與AD患者淀粉樣變的發(fā)生。
[Abstract]:Objective to investigate whether A 尾 -amyloidosis is present in the platelets of healthy people. The platelet count was determined by Sysmex-200 blood cell analyzer, the platelet distribution width was determined, the mean platelet volume, platelet specific volume and the ratio of large platelets were determined. The platelets were further purified, and the platelets were divided into two parts. Part of it is used to extract platelet membrane proteins, The other part was the detection of amyloidosis on platelet membrane with the fluorescence probe of sulfur Thunb Th Th. The immunoreaction of anti-A 尾 antibody in detecting amyloidosis prepeptide. Western blotting was used to observe the molecular polymerization of A 尾 protein. Almost all of the plates showed positive reaction. Further anti-A 尾 immunocytochemical assay found that the protein extracted from platelets contained macromolecular A 尾 polymer by superposition of green fluorescence and A 尾 antibody reaction. Conclusion\\\; There is A 尾 -amyloidosis in human platelets. It may be involved in the development of amyloidosis in AD patients.
