【摘要】：目的：探討單次鼻腔給予胰島素對(duì)異丙酚引起的tau蛋白過度磷酸化的影響及機(jī)制。 方法：SD大鼠，雄性,8周，給予以下處理：鼻腔給予胰島素（3ml:300IU）或生理鹽水42μl,半小時(shí)后腹腔注射異丙酚150mg/kg或等體積脂肪乳，一個(gè)小時(shí)后頸椎脫臼法處死，取雙側(cè)海馬組織，通過Westernblot檢測(cè)tau蛋白Thr205位點(diǎn)的磷酸化，GSK-3β、AKT、ERK、PP2A活性的變化。 結(jié)果：異丙酚引起tau蛋白Thr205位點(diǎn)的過度磷酸化,這種變化與磷酸激酶GSK-3β、AKT、ERK的活性變化關(guān)系不大，而主要可能由于磷酸酶PP2A總量下降所致。單次鼻腔給予胰島素并沒有逆轉(zhuǎn)異丙酚引起tau蛋白Thr205位點(diǎn)的過度磷酸化。 結(jié)論：單次鼻腔給予胰島素并不能對(duì)異丙酚引起的相關(guān)異常變化產(chǎn)生顯著的保護(hù)作用，，采用胰島素作為術(shù)后認(rèn)知功能障礙的保護(hù)措施尚需進(jìn)一步研究。
[Abstract]:Aim: to investigate the effect and mechanism of single nasal insulin administration on propofol induced hyperphosphorylation of tau protein. Methods male SD rats were given the following treatment for 8 weeks: nasal administration of insulin (3ml: 300 IUU) or normal saline (42 渭 l), intraperitoneal injection of propofol 150 mg / kg or fat emulsion of equal volume half an hour later, cervical spine dislocations were performed one hour later, and bilateral hippocampal tissues were taken. The activity of PP2A at Thr205 site of tau protein was detected by Westernblot. Results: propofol induced excessive phosphorylation of Thr205 site of tau protein. This change was not related to the activity of phosphokinase GSK-3 尾 -AKTERK. However, the decrease in the total amount of phosphatase PP2A may be the main reason. The single nasal insulin administration did not reverse the excessive phosphorylation of the Thr205 site of tau protein induced by propofol. Conclusion: single nasal administration of insulin can not significantly protect propofol related abnormal changes. The use of insulin as a protective measure for postoperative cognitive dysfunction needs further study.
