瑞舒伐他汀聯(lián)合臍血間充質(zhì)干細(xì)胞移植改善急性心肌梗死后心功能
[Abstract]:Background: in recent years, the regeneration of myocardial cells in infarcted area induced by stem cell transplantation has become a hot new therapeutic method for the treatment of cardiac insufficiency after acute myocardial infarction. Aim: to observe the effect of resuvastatin combined with umbilical cord blood mesenchymal stem cell transplantation on cardiac function after acute myocardial infarction in rats. Methods: 45 SD rats were randomly divided into three groups: model group, umbilical cord blood mesenchymal stem cell transplantation group and combined group with 15 rats in each group. On the 7th day after coronary artery ligation, the umbilical cord blood mesenchymal stem cell transplantation group and the combined group were injected with 300 渭 L (15.0 脳 108 cells) of mesenchymal stem cell suspension via tail vein. The model group was injected with the same dose of LG-DMEM culture medium. The combined group was given resuvastatin 1 mg/kg intragastrically for 28 days (from the first day after operation), the umbilical cord blood mesenchymal stem cell transplantation group and the model group were given the same dose of normal saline simultaneously. The improvement of cardiac function was detected by color Doppler ultrasound and the plasma Lp-PLA2 level was detected by enzyme-linked immunosorbent assay (Elisa) 5 weeks after the model (4 weeks of cell transplantation). The pathological changes of myocardium were observed by hematoxylin-eosin staining. The expression of heat shock protein 70 in myocardial infarction was detected by Western blot. Results and conclusion the left ventricular ejection fraction (LVEF) of rats in the combined group of 1: 1, The left ventricular end-systolic pressure was significantly higher than that of cord blood mesenchymal stem cell transplantation group (P0.05), the umbilical cord blood mesenchymal stem cell transplantation group was significantly higher than the model group (P0.05), and the left ventricular end-diastolic pressure of the combined group was significantly lower than that of the umbilical cord blood mesenchymal stem cell transplantation group (P0.05). (P0.05), the level of Lp-PLA2 in the umbilical cord blood mesenchymal stem cell transplantation group was significantly lower than that in the model group (P0.05). The level of Lp-PLA2 in the combined group was significantly lower than that in the cord blood mesenchymal stem cell transplantation group (P0.05), and it was significantly lower in the umbilical cord blood mesenchymal stem cell transplantation group than in the model group (P0.05). The number of cells in the model group was higher than that in the model group. In cord blood mesenchymal stem cell transplantation group, the expression of HSP70 protein in myocardial infarction was significantly increased in the combined group, and there was significant difference among the groups (P0.05). Resuvastatin combined with umbilical cord blood mesenchymal stem cell transplantation can improve cardiac function of myocardial infarction rats.
