【摘要】：背景:近年來(lái)通過(guò)干細(xì)胞移植誘導(dǎo)梗死區(qū)心肌細(xì)胞的再生用以治療急性心肌梗死后心功能不全,成為較為熱議的一種新的治療手段。目的:觀察瑞舒伐他汀聯(lián)合臍血間充質(zhì)干細(xì)胞移植對(duì)大鼠急性心肌梗死后心功能的改善作用。方法:選取45只SD大鼠采用結(jié)扎左冠狀動(dòng)脈前降支方法制備心肌梗死模型,隨機(jī)分為模型組、臍血間充質(zhì)干細(xì)胞移植組和聯(lián)合組,每組15只。大鼠冠脈結(jié)扎后第7天,臍血間充質(zhì)干細(xì)胞移植組及聯(lián)合組經(jīng)尾靜脈注射臍血間充質(zhì)干細(xì)胞懸液300μL(15.0×108個(gè)細(xì)胞),模型組注射相同劑量LG-DMEM培養(yǎng)液,聯(lián)合組給予瑞舒伐他汀1 mg/kg灌胃×28 d(從術(shù)后第1天開(kāi)始灌胃,1次/d),臍血間充質(zhì)干細(xì)胞移植組及模型組同步灌胃相同劑量的生理鹽水。造模后5周(細(xì)胞移植4周)采用彩色多普勒超聲儀檢測(cè)心功能改善情況,酶聯(lián)免疫吸附法檢測(cè)血漿Lp-PLA2水平,蘇木精-伊紅染色觀察心肌組織病理變化;Western blot方法檢測(cè)心肌梗死區(qū)熱休克蛋白70表達(dá)。結(jié)果與結(jié)論:1聯(lián)合組大鼠的左心室射血分?jǐn)?shù)、左心室收縮末壓明顯高于臍血間充質(zhì)干細(xì)胞移植組(P0.05),臍血間充質(zhì)干細(xì)胞移植組明顯高于模型組(P0.05);聯(lián)合組大鼠的左心室舒張末壓明顯低于臍血間充質(zhì)干細(xì)胞移植組(P0.05),臍血間充質(zhì)干細(xì)胞移植組明顯低于模型組(P0.05);2聯(lián)合組Lp-PLA2水平明顯低于臍血間充質(zhì)干細(xì)胞移植組(P0.05),臍血間充質(zhì)干細(xì)胞移植組明顯低于模型組(P0.05);3聯(lián)合組梗死區(qū)的心肌細(xì)胞數(shù)量較模型組、臍血間充質(zhì)干細(xì)胞移植組明顯增加;4心肌梗死區(qū)HSP70蛋白表達(dá)在聯(lián)合組明顯升高,各組間的表達(dá)差異有顯著性意義(P0.05);5結(jié)果表明,瑞舒伐他汀聯(lián)合臍血間充質(zhì)干細(xì)胞移植干預(yù)后能確切改善心肌梗死大鼠心功能。
[Abstract]:Background: in recent years, the regeneration of myocardial cells in infarcted area induced by stem cell transplantation has become a hot new therapeutic method for the treatment of cardiac insufficiency after acute myocardial infarction. Aim: to observe the effect of resuvastatin combined with umbilical cord blood mesenchymal stem cell transplantation on cardiac function after acute myocardial infarction in rats. Methods: 45 SD rats were randomly divided into three groups: model group, umbilical cord blood mesenchymal stem cell transplantation group and combined group with 15 rats in each group. On the 7th day after coronary artery ligation, the umbilical cord blood mesenchymal stem cell transplantation group and the combined group were injected with 300 渭 L (15.0 脳 108 cells) of mesenchymal stem cell suspension via tail vein. The model group was injected with the same dose of LG-DMEM culture medium. The combined group was given resuvastatin 1 mg/kg intragastrically for 28 days (from the first day after operation), the umbilical cord blood mesenchymal stem cell transplantation group and the model group were given the same dose of normal saline simultaneously. The improvement of cardiac function was detected by color Doppler ultrasound and the plasma Lp-PLA2 level was detected by enzyme-linked immunosorbent assay (Elisa) 5 weeks after the model (4 weeks of cell transplantation). The pathological changes of myocardium were observed by hematoxylin-eosin staining. The expression of heat shock protein 70 in myocardial infarction was detected by Western blot. Results and conclusion the left ventricular ejection fraction (LVEF) of rats in the combined group of 1: 1, The left ventricular end-systolic pressure was significantly higher than that of cord blood mesenchymal stem cell transplantation group (P0.05), the umbilical cord blood mesenchymal stem cell transplantation group was significantly higher than the model group (P0.05), and the left ventricular end-diastolic pressure of the combined group was significantly lower than that of the umbilical cord blood mesenchymal stem cell transplantation group (P0.05). (P0.05), the level of Lp-PLA2 in the umbilical cord blood mesenchymal stem cell transplantation group was significantly lower than that in the model group (P0.05). The level of Lp-PLA2 in the combined group was significantly lower than that in the cord blood mesenchymal stem cell transplantation group (P0.05), and it was significantly lower in the umbilical cord blood mesenchymal stem cell transplantation group than in the model group (P0.05). The number of cells in the model group was higher than that in the model group. In cord blood mesenchymal stem cell transplantation group, the expression of HSP70 protein in myocardial infarction was significantly increased in the combined group, and there was significant difference among the groups (P0.05). Resuvastatin combined with umbilical cord blood mesenchymal stem cell transplantation can improve cardiac function of myocardial infarction rats.
