復方蘋果酸鈉對失血性休克大鼠小腸黏膜細胞凋亡的影響

【摘要】:目的觀察復方蘋果酸鈉對失血性休克大鼠小腸黏膜細胞凋亡的影響。方法雄性SD大鼠48只,體重280~320g,隨機分為假手術(shù)組(SS組)、生理鹽水組(NS組)、復方乳酸鈉組(RL組)和復方蘋果酸鈉組(RM組),每組12只。SS組為空白對照組,其余三組制作失血性休克模型,放血至MAP降為(40±5)mm Hg,休克60min后進行對應液體復蘇,復蘇后3h處死大鼠。檢測小腸黏膜中B淋巴細胞瘤-2基因(Bcl-2)表達量和Bcl-2相關(guān)蛋白(Bax)含量,采用TUNEL法檢測黏膜細胞凋亡情況,計算凋亡指數(shù)。檢測小腸組織中超氧化物歧化酶(SOD)活性和丙二醛(MDA)濃度。取小腸黏膜做病理切片,觀察病理改變。結(jié)果 NS、RL、RM組Bcl-2表達量明顯低于SS組,Bax含量及凋亡指數(shù)明顯高于SS組(P0.05);RM組Bcl-2表達量明顯高于NS組和RL組,Bax含量及凋亡指數(shù)明顯低于NS組和RL組(P0.05)。NS、RL、RM組SOD活性明顯低于SS組,MDA濃度明顯高于SS組(P0.05);RM組SOD活性明顯高于NS組和RL組,MDA濃度明顯低于NS組和RL組(P0.05)。病理組織切片顯示RM組小腸黏膜損傷輕于NS組和RL組。結(jié)論復方蘋果酸鈉對于失血性休克大鼠小腸黏膜具有良好的抗凋亡作用,為臨床晶體液的應用帶來新的選擇。
[Abstract]:Objective to observe the effect of compound malate sodium on apoptosis of intestinal mucosal cells in hemorrhagic shock rats. Methods Forty-eight male SD rats, weighing 280 ~ 320g, were randomly divided into three groups: sham operation group (SS group), saline group (NS group), compound sodium lactate group (RL group) and compound malate sodium group (RM group). The model of hemorrhagic shock was made in the other three groups, and the bleeding to map was reduced to (40 鹵5) mm Hg. The corresponding fluid resuscitation was performed after shock and 60min, and the rats were killed 3 hours after resuscitation. The expression of B lymphocyte tumor 2 gene (Bcl-2) and Bcl-2 associated protein (Bax) in small intestinal mucosa were detected. Tunel method was used to detect the apoptosis of mucosal cells and the apoptotic index was calculated. The activity of superoxide dismutase (SOD) and the concentration of malondialdehyde (MDA) in small intestine were measured. The intestinal mucosa was taken as pathological section and the pathological changes were observed. Results the expression of Bcl-2 in RM group was significantly lower than that in SS group (P0.05). The expression of Bcl-2 in RM group was significantly higher than that in NS group and RL group, and the apoptotic index was significantly lower than that in NS group and RL group (P0.05) .NSL RLM group was significantly lower than that in NS group and RL group (P0.05). The activity of SOD in RM group was significantly lower than that in RL group (P0.05). The concentration of MDA in SS group was significantly higher than that in SS group (P0.05). The activity of SOD in RM group was significantly higher than that in NS group and RL group (P0.05). Pathological sections showed that intestinal mucosal damage in RM group was less than that in NS and RL groups. Conclusion compound sodium malate has a good antiapoptotic effect on the intestinal mucosa of hemorrhagic shock rats, which brings a new choice for the application of clinical lens fluid.
