腦損傷與他克莫司促進(jìn)周?chē)窠?jīng)再生的對(duì)比研究

【摘要】:目的通過(guò)比較腦損傷、他克莫司促進(jìn)大鼠坐骨神經(jīng)再生修復(fù)的機(jī)制,探索腦損傷促進(jìn)周?chē)窠?jīng)損傷修復(fù)機(jī)制。方法實(shí)驗(yàn)動(dòng)物隨機(jī)分為4組,顱腦損傷合并坐骨神經(jīng)損傷模型(A組),單純坐骨神經(jīng)損傷模型(B組),造模后分別給予A1組他克莫司、A2組等量生理鹽水、B1組他克莫司、B2組等量生理鹽水,2周。分別于術(shù)后第4、8、12周,進(jìn)行測(cè)定坐骨神經(jīng)指數(shù)、腓腸肌恢復(fù)率、肌肉Masson染色、神經(jīng)組織蘇木素-伊紅染色、Masson染色、甲苯胺藍(lán)染色,術(shù)后8、12周測(cè)定坐骨神經(jīng)干的動(dòng)作電位幅值恢復(fù)率、辣根過(guò)氧化物示蹤檢測(cè)。結(jié)果通過(guò)實(shí)驗(yàn)比較,腦損傷聯(lián)合應(yīng)用他克莫司促進(jìn)周?chē)窠?jīng)損傷的修復(fù)效果坐骨神經(jīng)指數(shù)、神經(jīng)干的動(dòng)作電位幅值恢復(fù)率、腓腸肌恢復(fù)率、肌肉Masson染色、神經(jīng)組織蘇木素-伊紅染色、Masson染色、甲苯胺藍(lán)染色、辣根過(guò)氧化物示蹤檢測(cè)優(yōu)于其它單獨(dú)因素的作用。結(jié)論腦損傷與他克莫司促進(jìn)周?chē)窠?jīng)損傷的修復(fù)的機(jī)制不同,其促進(jìn)周?chē)窠?jīng)損傷修復(fù)的機(jī)制仍需進(jìn)一步探討。
[Abstract]:Objective to compare the mechanism of brain injury and tacrolimus promoting sciatic nerve regeneration and repair in rats and explore the mechanism of brain injury promoting peripheral nerve injury. Methods the experimental animals were randomly divided into 4 groups. The model of craniocerebral injury combined with sciatic nerve injury (group A) and the model of simple sciatic nerve injury (group B) were treated with tacrolimide A2 group (A1 group) and tacrolimus B2 group (B 1 group) respectively for 2 weeks. The sciatic nerve index, gastrocnemius recovery rate, muscle Masson staining, hematoxylin-eosin staining and toluidine blue staining were measured at the 8th week after operation. The recovery rate of action potential amplitude of sciatic nerve trunk and horseradish peroxide tracer were measured 812 weeks after operation. Results the effects of tacrolimus combined with brain injury on the repair of peripheral nerve injury were compared. The sciatic nerve index, the recovery rate of action potential amplitude of nerve trunk, the recovery rate of gastrocnemius muscle and Masson staining of muscle were observed. The effects of hematoxylin-eosin staining Masson staining toluidine blue staining horseradish peroxide tracer detection were superior to those of other single factors. Conclusion the mechanism of brain injury is different from that of tacrolimus in promoting the repair of peripheral nerve injury.
