KLF4在創(chuàng)傷新生血管內(nèi)皮細(xì)胞中的表達(dá)和對(duì)血管內(nèi)皮細(xì)胞功能的影響

【摘要】:目的 觀察皮膚創(chuàng)傷愈合過程中KLF4(Kruppel-LikeFactor4)在新生血管內(nèi)皮細(xì)胞中的表達(dá)及在體外對(duì)血管內(nèi)皮細(xì)胞增殖、遷移及管腔形成能力的影響。 方法 用24只大鼠作皮膚創(chuàng)傷模型,分別于創(chuàng)傷后即刻、2、4、6、8、10、12、14d處死并取創(chuàng)緣組織,應(yīng)用免疫組織化學(xué)技術(shù)和圖像分析技術(shù)觀察創(chuàng)傷模型大鼠在不同時(shí)間點(diǎn)皮膚創(chuàng)緣組織新生血管內(nèi)皮細(xì)胞中KLF4的表達(dá)情況;在體外培養(yǎng)人臍靜脈內(nèi)皮細(xì)胞(HUVECs),,用KLF4小干擾RNA(siRNA)轉(zhuǎn)染人臍靜脈內(nèi)皮細(xì)胞(HUVECs)48h后,應(yīng)用CellCountingKit-8(CCK-8)、細(xì)胞劃痕愈合實(shí)驗(yàn)以及體外血管形成實(shí)驗(yàn)分別檢測(cè)轉(zhuǎn)染前后HUVECs的增殖、遷移以及管腔形成能力的變化。 結(jié)果 與對(duì)照組相比,KLF4在肉芽組織新生血管內(nèi)皮細(xì)胞中有表達(dá);轉(zhuǎn)染siRNA下調(diào)KLF4的表達(dá)后,HUVECs的增殖、遷移及管腔形成能力較對(duì)照組均明顯下降,轉(zhuǎn)染前HUVECs的吸光度(A)值、遷移距離和成管環(huán)數(shù)分別為1.396±0.025、497±6.2、32±0.7,轉(zhuǎn)染后為0.948±0.043、206±5.6、12±0.7(P0.05)。 結(jié)論 本研究證實(shí)KLF4參與皮膚創(chuàng)傷修復(fù)過程,對(duì)皮膚創(chuàng)傷愈合過程中新生血管
[Abstract]:Objective to observe the expression of KLF4- Kruppel-LikeFactor4 in neovascularization endothelial cells and its effect on proliferation, migration and lumen formation of vascular endothelial cells in vitro. Methods 24 rats were used as skin trauma models. The wound tissue was killed immediately after trauma and the expression of KLF4 was observed by immunohistochemistry and image analysis at different time points in the neovascularization endothelial cells of the wound model rats. Human umbilical vein endothelial cells (HUVECs) were cultured in vitro. HUVECs were transfected with KLF4 small interference RNA-siRNAs for 48 hours. Cell counting Kit-8CCK-8 was used to detect the proliferation of HUVECs before and after transfection. Results compared with the control group, KLF4 was expressed in neovascularization endothelial cells, and the proliferation, migration and lumen formation of HUVECs were significantly decreased after transfection of siRNA down-regulated the expression of KLF4. Before transfection, the absorbance of HUVECs, the migration distance and the number of tubular rings were 1.396 鹵0.025497 鹵6.2, 32 鹵0.7, and 0.948 鹵0.043206 鹵5.6, 12 鹵0.7, respectively. Conclusion this study confirmed that KLF4 is involved in the repair of skin trauma and the neovascularization in the healing process of skin trauma.
