糞便細(xì)菌移植對(duì)肝性腦病大鼠肝功能及血氨的影響

【摘要】:目的建立大鼠肝性腦病及FMT細(xì)菌模型,確定糞便細(xì)菌移植對(duì)肝性腦病大鼠肝功能及血氨的影響,初步了解糞便細(xì)菌移植(fecal microbiota transplantation,FMT)的臨床意義。方法選擇32只SD大鼠,采用CCL4+酒精的方法建立大鼠肝性腦病模型及腸道埋管,分為4組(A組:正常對(duì)照組;B組:肝性腦病模型組;C組:益生菌組;D組:FMT組),對(duì)C組進(jìn)行益生菌移植,200μL/d(200μL益生菌溶液中含益生菌2.7×10~9),持續(xù)2周;對(duì)D組進(jìn)行FMT移植,200μL/d(200μL糞便細(xì)菌溶液中含糞便細(xì)菌2.7×10~9),持續(xù)2周。記錄大鼠一般生存情況及大便情況,對(duì)移植前后體重進(jìn)行比較,測(cè)定移植前、后肝功能(ALT、AST、ALB、TB及DB)和門靜脈及尾靜脈血氨。結(jié)果所有實(shí)驗(yàn)大鼠均未出現(xiàn)死亡;糞便細(xì)菌移植后肝性腦病大鼠體重移植前明顯增加[D組移植前(225.18±14.81)g,移植后2周(266.83±33.40)g,P0.05];移植后大鼠肝功能較前明顯好轉(zhuǎn)[移植后2周ALT:D組與B組分別為(610.49±4.98)U/L和(990.07±4.80)U/L,P0.05;AST:D組與B組分別為(72.46±4.42)U/L和(101.58±2.19)U/L,P0.05;ALB:D組與B組比較ALB明顯上升,分別為(24.48±0.12)g/L和(17.53±0.53)g/L,P0.05;TB:D組為(13.45±0.77)μmol/L,B組為(27.20±0.77)μmol/L,P0.05;DB:D組為(10.04±0.09)μmol/L,B組為(16.04±0.51)μmol/L,P0.05];門靜脈及尾靜脈血氨較移植前明顯降低[D組門靜脈及尾靜脈血氨分別為(26.26±0.30)μmol/L和(23.10±0.35)μmol/L,B組分別為(36.28±1.32)μmol/L和(32.90±0.35)μmol/L,P0.05]。結(jié)論對(duì)大鼠進(jìn)行人體糞便細(xì)菌移植是可行的;糞便細(xì)菌移植可改善肝性腦病大鼠的肝功能,降低血氨。
[Abstract]:Objective to establish a rat model of hepatic encephalopathy and FMT bacteria, to determine the effect of fecal bacterial transplantation on liver function and blood ammonia in rats with hepatic encephalopathy, and to understand the clinical significance of fecal bacterial transplantation. Methods 32 Sprague-Dawley rats were selected to establish hepatic encephalopathy model and intestinal canal embedding by CCL4 alcohol. The rats were divided into 4 groups: normal control group, group B: liver encephalopathy model group, group C: probiotics group D: FMT group, probiotics group C containing 2. 7 脳 10 ~ 9 probiotics in probiotic solution of 200 渭 L/d(200 渭 L of probiotics for 2 weeks.
