高同型半胱氨酸血癥對急性ST段抬高型心肌梗死患者淋巴細胞KV1.3-CaN-NFAT信號通路的影響

【摘要】:目的觀察不同水平高同型半胱氨酸血癥(Hhcy)對急性ST段抬高型心肌梗死(STEMI)患者淋巴細胞內(nèi)電壓門控性鉀離子通道(KV1.3通道)-鈣調(diào)神經(jīng)磷酸酶(CaN)-活化T細胞核因子(NFAT)-炎性因子(IL-6、TNF-α)mRNA及蛋白表達水平的影響。方法入選STEMI患者90例,根據(jù)血漿Hcy水平分為3組:實驗A組(STEMI組,Hcy15μmol/L,n=30)、實驗B組(STEMI+輕度Hhcy組,Hcy 15~30μmol/L,n=30)、實驗C組(STEMI+中度Hhcy組,Hcy30μmol/L,n=30),另選取健康體檢者30例為對照組。抽取血標(biāo)本后使用全自動生化儀測定血漿Hcy濃度,密度梯度離心法提取淋巴細胞,RT-PCR檢測KV1.3、CnAα、NFAT1、IL-6、TNF-αmRNA的表達,Western blot檢測KV1.3、CnAα、NFAT1蛋白的表達。結(jié)果各實驗組較對照組KV1.3、CnAα和NFAT1的mRNA和蛋白表達水平升高(P0.05或P0.01),各實驗組間兩兩比較顯示,實驗B組較實驗A組表達水平升高(P0.05或P0.01),實驗C組較實驗B組和實驗A組表達水平升高(P0.05或P0.01)。各實驗組IL-6和TNF-α的mRNA表達水平高于對照組(P0.01),各實驗組間兩兩比較顯示,實驗B組較實驗A組表達水平升高(P0.01),實驗C組較實驗B組和實驗A組表達水平升高(P0.01)。實驗組血漿Hcy水平與KV1.3mRNA和蛋白表達之間呈正相關(guān),相關(guān)系數(shù)分別為r=0.503(P=0.000)、r=0.726(P=0.000)。結(jié)論血漿Hcy濃度水平越高,STEMI患者淋巴細胞內(nèi)KV1.3-CnAα-NFAT1的mRNA和蛋白表達水平及炎性因子IL-6、TNF-α的mRNA表達水平越高,這一信號通路可能是Hcy加重STEMI炎癥反應(yīng)的機制之一。
[Abstract]:Objective to observe the effect of different levels of hyperhomocysteinemia (Hhcyia) on the intracellular voltage-gated potassium channel (KV 1.3 channel) and calmodulating neuronal phosphatase (CaN) -activated T cell nuclear factor (NFATAT-T) in patients with acute ST-segment elevation myocardial infarction (STEMI). The expression level of TNF- 偽 mRNA and protein was influenced by IL-6 and TNF- 偽. Methods 90 patients with STEMI were divided into three groups according to their plasma Hcy levels: group A: Hcy15 渭 mol 路L ~ (-1) 路L ~ (-1) Hcy, group B (n = 30) with mild Hhcy, group C (n = 30) with moderate Hhcy, group C (n = 30) with Hcy30 渭 mol 路L ~ (-1) 路L ~ (-1), and control group (n = 30) with normal control group (n = 30). Blood samples were collected and plasma Hcy concentration was measured by automatic biochemical instrument. RT-PCR was used to detect the expression of IL-6 TNF- 偽 mRNA in lymphocytes extracted by density gradient centrifugation. Results the expression levels of mRNA and protein of CnA 偽 and NFAT1 in each experimental group were significantly higher than those in the control group (P0.05 or P0.01). Compared with group A, the level of expression in group B was higher than that in group A (P0.05 or P0.01), and the level of expression in group C was higher than that in group B and group A (P0.05 or P0.01). The expression level of IL-6 and TNF- 偽 in each experimental group was higher than that in control group (P 0.01). The expression level of IL-6 and TNF- 偽 in experimental group B was higher than that in group A, and the expression level of mRNA in group C was higher than that in group B and group A. the expression level of TNF- 偽 in experimental group was higher than that in group B and group A. There was a positive correlation between the plasma Hcy level and the expression of KV1.3mRNA and protein in the experimental group. Conclusion the higher the level of plasma Hcy, the higher the mRNA and protein expression of KV1.3-CnA 偽 -NFAT1 and the higher the mRNA expression of IL-6 TNF- 偽 in the lymphocytes of patients with STEMI. This signaling pathway may be one of the mechanisms of Hcy exacerbating the inflammatory response of STEMI.
