【摘要】：目的研究選擇性環(huán)氧合酶-2抑制劑——塞來昔布對(duì)大鼠顱腦創(chuàng)傷后凋亡蛋白Bcl-2和Bax表達(dá)的影響。方法按照體重將SD大鼠隨機(jī)分為4組:實(shí)驗(yàn)組、模型組、假手術(shù)組和對(duì)照組,實(shí)驗(yàn)組和模型組構(gòu)建大鼠閉合性顱腦創(chuàng)傷模型。在顱腦損傷模型構(gòu)建后,實(shí)驗(yàn)組立即經(jīng)腹腔注入塞來昔布250mg·kg-1;模型組、假手術(shù)組和對(duì)照組給于同等量的0.9%Na Cl腹腔注射。用免疫印跡法和免疫組織化學(xué)染色法檢測凋亡蛋白Bcl-2和Bax的表達(dá)。結(jié)果實(shí)驗(yàn)組、模型組、假手術(shù)組和對(duì)照組的各組Bcl-2蛋白表達(dá)量分別為68.75±0.62,45.40±0.36,81.08±0.65,78.12±0.05,與假手術(shù)組和對(duì)照組比較,模型組Bcl-2蛋白表達(dá)量明顯降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05);與模型組比較,實(shí)驗(yàn)組Bcl-2蛋白表達(dá)量明顯增加,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。實(shí)驗(yàn)組、模型組、假手術(shù)組和對(duì)照組的各組Bax蛋白表達(dá)量分別為68.28±0.35,92.48±0.16,65.06±0.63,63.15±0.10,與假手術(shù)組和對(duì)照組比較,模型組Bax蛋白表達(dá)量明顯增加,差異有統(tǒng)計(jì)學(xué)意義(P0.05);與模型組相比較,實(shí)驗(yàn)組Bax蛋白表達(dá)量明顯降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論塞來昔布可通過增加腦組織Bcl-2的表達(dá)、降低Bax的表達(dá)和抑制腦損傷后神經(jīng)細(xì)胞的凋亡,對(duì)大鼠顱腦創(chuàng)傷具有保護(hù)作用。
[Abstract]:Objective to study the effect of selective cyclooxygenase -2 inhibitor - celecoxib on the expression of apoptotic protein Bcl-2 and Bax in rats after craniocerebral trauma. Methods SD rats were randomly divided into 4 groups according to body weight: experimental group, model group, sham operation group and control group, experimental group and model group constructed rat model of closed craniocerebral trauma. The model of brain injury in the experimental group and model group. After the construction, the experimental group was immediately injected with celecoxib 250mg kg-1; the model group, the sham operation group and the control group were given the same amount of 0.9%Na Cl intraperitoneally. The expression of Bcl-2 and Bax of apoptotic protein was detected by immunoblotting and immunohistochemistry. Results of the experimental group, model group, sham operation group and control group Bcl-2 protein table The amount of the expression was 68.75 + 0.62,45.40 + 0.36,81.08 + 0.65,78.12 + 0.05 respectively. Compared with the sham operation group and the control group, the expression of Bcl-2 protein in the model group decreased significantly (P0.05). The experimental group, the model group, the sham operation group and the experimental group were statistically significant (P0.05). The expression of Bax protein in the control group was 68.28 + 0.35,92.48 + 0.16,65.06 + 0.63,63.15 + 0.10 respectively. Compared with the sham operation group and the control group, the expression of Bax protein in the model group was significantly increased, the difference was statistically significant (P0.05). Compared with the model group, the expression of Bax protein in the experimental group was significantly reduced, the difference was statistically significant (P0.05). Celecoxib can increase the expression of Bcl-2 in brain tissue, reduce the expression of Bax and inhibit the apoptosis of nerve cells after brain injury. It has protective effect on brain trauma in rats.
