中性粒細胞彈性蛋白酶抑制劑對顱腦損傷大鼠腦組織的保護作用

【摘要】:目的探討中性粒細胞彈性蛋白酶(NE)抑制劑西維來司鈉對顱腦損傷(TBI)大鼠腦組織的保護作用及機制。方法 SD大鼠70只,隨機分為對照組(n=10)、TBI組(n=30)和干預組(n=30),TBI組和干預組又分為24、72、168 h亞組。用液壓打擊方法制備TBI模型,TBI組術(shù)后立即腹腔注射生理鹽水,干預組術(shù)后立即腹腔注射西維來司鈉。于術(shù)后24、72和168 h取挫傷側(cè)腦組織標本,測定NE、超氧化物歧化酶(SOD)、丙二醛(MDA)和髓過氧化物酶(MPO)活性和含量。結(jié)果與對照組相比,TBI組腦組織NE、MDA含量和MPO活性明顯增高(P0.05),SOD活性明顯降低(P0.05);與TBI組相比,干預組腦組織NE、MDA含量和MPO活性明顯降低(P0.05),SOD活性明顯增高(P0.05)。結(jié)論西維來司鈉可以通過抑制NE活性,抑制中性粒細胞活化和浸潤,減少自由基的產(chǎn)生及脂質(zhì)過氧化,保護神經(jīng)細胞和內(nèi)皮細胞的膜性結(jié)構(gòu)和功能的完整,對TBI大鼠腦組織具有保護作用。
[Abstract]:Objective to investigate the protective effect and mechanism of neutrophil elastase (NEN) inhibitor sivelestat sodium on brain tissue of TBI rats with craniocerebral injury. Methods 70 SD rats were randomly divided into control group (n = 10), control group (n = 30), intervention group (n = 30) and intervention group (n = 72 168 h). The TBI model was made by hydraulic beating. The rats in the intervention group received intraperitoneal injection of normal saline immediately after operation, and the intervention group received intraperitoneal injection of sivelestat sodium immediately after operation. The activity and content of NE, SOD, MDA and MPO in contused brain tissue were measured at 24 h, 72 h and 168 h after operation. Results compared with the control group, the content of MPO and the content of MPO in brain tissue of TBI group were significantly higher than those of the control group, and the activity of P0.05sSOD in brain tissue of the intervention group was significantly lower than that of the control group, and the content of MPO in the brain tissue of the intervention group were significantly lower than that of the TBI group. The activity of MPO in the brain tissue of the intervention group was significantly increased than that of the control group. Conclusion sivelestat sodium can inhibit NE activity, neutrophil activation and infiltration, reduce free radical production and lipid peroxidation, and protect the integrity of membrane structure and function of nerve cells and endothelial cells. It has protective effect on brain tissue of TBI rats.
