大鼠肝臟炎癥環(huán)境下肝卵圓細(xì)胞增殖模型的建立及評(píng)價(jià)

【摘要】:目的:建立肝臟炎癥環(huán)境下肝卵圓細(xì)胞(HOC)的增殖模型,初步觀察肝臟炎癥環(huán)境下肝卵圓細(xì)胞的部分干細(xì)胞生物學(xué)特性方法:將55只雄性SD大鼠隨機(jī)分為正常對(duì)照組、慢性肝炎模型組和肝炎+2-AAF模型組、2-AAF/PH模型組。采用CCL4復(fù)合法制備慢性肝炎動(dòng)物模型;以CCL4復(fù)合法為基礎(chǔ),灌胃2-乙酰氨基芴(2-AAF)制備炎癥環(huán)境下肝卵圓細(xì)胞增殖模型;肝2/3切除并術(shù)后灌胃2-AAF制備非炎癥環(huán)境下HOC增殖模型。HE染色及VG膠原染色觀察肝病理組織學(xué)改變,免疫組織化學(xué)法觀察EPCAM和Thy-1的表達(dá)。結(jié)果:正常對(duì)照組大鼠肝組織中未見肝卵圓細(xì)胞;肝炎癥模型組可見少量的肝卵圓細(xì)胞增殖;肝炎癥+2-AAF模型組可見明顯的肝卵圓細(xì)胞增殖,其中以8周的肝組織中最顯著。與2-AAF/PH非炎癥模型組比較,肝炎癥環(huán)境中肝卵圓細(xì)胞高表達(dá)Thy-1。結(jié)論:CCL4復(fù)合法結(jié)合2-AAF灌胃可構(gòu)建較為穩(wěn)定的炎癥環(huán)境下HOC增殖模型;炎癥環(huán)境是HOC表現(xiàn)出肝癌干細(xì)胞部分生物學(xué)特性的原因之一。
[Abstract]:Objective: to establish the proliferative model of hepatic oval cell (Hoca) in the environment of liver inflammation, and to observe the biological characteristics of some stem cells in hepatic oval cells. Methods: 55 male Sprague-Dawley rats were randomly divided into normal control group.Chronic hepatitis model group and hepatitis 2-AAF model group 2-AAFP-PH model group.The animal model of chronic hepatitis was established by CCL4 compound method, and the proliferation model of hepatic oval cells was established by intragastric administration of 2-acetylaminofluorene (2-AAFF) on the basis of CCL4 compound method.HOC proliferation model was established by 2 / 3 resection of liver and intragastric administration of 2-AAF after operation. He staining and VG collagen staining were used to observe the histopathological changes of liver and the expression of EPCAM and Thy-1 were observed by immunohistochemical method.Results: no hepatic oval cells were found in the normal control group, a small number of hepatic oval cells proliferated in the liver inflammation model group, and obvious hepatic oval cell proliferation was observed in the 2-AAF model group, especially in the liver tissue at 8 weeks.The expression of Thy-1 in hepatic oval cells was higher than that in 2-AAF/PH non-inflammatory model group.Conclusion the HOC proliferation model can be constructed by the combination of 2-AAF with the combined method of 10% CCL4, and the inflammatory environment is one of the reasons why HOC shows some biological characteristics of HCC stem cells.
