【摘要】：目的 研究三磷酸腺苷結(jié)合盒轉(zhuǎn)運子A1基因R219K多態(tài)性與遼西地區(qū)漢族人群急性心肌梗死易感性及血脂水平的相關性。 方法 本研究選用遼西地區(qū)漢族人群作為研究對象，急性心肌梗死患者103例，正常對照114例。使用聚合酶鏈反應-限制性片段長度多態(tài)性技術對217例樣本進行基因分型，描述ABCA1基因R219K多態(tài)性基因型分布特征，，分析該位點與急性心肌梗死易感性的關聯(lián)。同時采用生化技術檢測研究對象的血脂水平，分析不同基因型與血脂水平的關系。 結(jié)果 1、ABCA1基因R219K多態(tài)性存在于遼西地區(qū)漢族人群中，該位點多態(tài)性有三種:RR型、RK型和KK型。在健康人群中RR型占29.8%，RK型占47.4%，KK型占22.8%。在急性心肌梗死組中RR型占43.7%，RK型占45.6%，KK型占10.7%。 2、ABCA1基因R219K多態(tài)性三種基因型頻率在急性心肌梗死組和對照組中分布差異有統(tǒng)計學意義(P0.05)；K等位基因在急性心肌梗死組和對照組中分布差異有統(tǒng)計學意義（P0.01）。 3、健康對照組中K等位基因攜帶者高密度脂蛋白膽固醇水平高于非K攜帶者，分布差異有統(tǒng)計學意義(P0.05)。對照組中甘油三酯、總膽固醇、低密度脂蛋白膽固醇水平在基因型亞組間的差異無統(tǒng)計學意義(P0.05)。 結(jié)論 ABCA1基因R219K位點多態(tài)性與急性心肌梗死有相關性；K等位基因可能是急性心肌梗死的保護基因。K等位基因攜帶者血漿高密度脂蛋白膽固醇水平高于非K攜帶者。
[Abstract]:Purpose. To study the relationship between R219K polymorphism of adenosine triphosphate binding cassette transporter A1 gene and susceptibility to acute myocardial infarction (AMI) and blood lipid level in Han population in western Liaoning province. Method. In this study, 103 patients with acute myocardial infarction (AMI) and 114 normal controls were enrolled in this study. 217 samples were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique. The distribution characteristics of R219K polymorphism genotype of ABCA1 gene were described, and the association between R219K polymorphism and susceptibility to acute myocardial infarction was analyzed. At the same time, the blood lipid level was detected by biochemical technique, and the relationship between different genotypes and serum lipid level was analyzed. Results. 1the R219K polymorphism of ABCA1 gene was found in Han population in western Liaoning, and there were three types of RK and KK in this locus. In the healthy population, the RR type was 29.8and the RK type was 47.4%. In the acute myocardial infarction group, the RR type was 43.7RK type and 45.6KK type, 10.7% in the acute myocardial infarction group. 2There was significant difference in the distribution of R219K polymorphism of ABCA1 gene between the acute myocardial infarction group and the control group. There was significant difference in the distribution of P0.05T allele between the acute myocardial infarction group and the control group. 3. The level of high density lipoprotein cholesterol in K allele carriers in healthy control group was higher than that in non K carriers, and the distribution difference was statistically significant (P 0.05). There was no significant difference in low density lipoprotein cholesterol level among genotypic subgroups (P 0.05). Conclusion. The polymorphism of R219K locus of ABCA1 gene is associated with acute myocardial infarction. It may be that the plasma high density lipoprotein cholesterol level in patients with acute myocardial infarction is higher than that in non-K carriers.
