劃痕損傷對(duì)大鼠大腦皮層微血管內(nèi)皮細(xì)胞NF-κB表達(dá)及繼發(fā)性死亡的影響

【摘要】:目的通過(guò)體外培養(yǎng)SD大鼠大腦皮層微血管內(nèi)皮細(xì)胞并制作劃痕損傷模型,觀察劃痕損傷對(duì)腦微血管內(nèi)皮細(xì)胞NF-κB表達(dá)及其自噬、凋亡和壞死的影響。方法體外培養(yǎng)SD大鼠大腦皮層微血管內(nèi)皮細(xì)胞并制作劃痕損傷模型,應(yīng)用NF-κB抑制劑吡咯醛二硫氨基甲酸(PDTC)干預(yù)劃痕損傷腦微血管內(nèi)皮細(xì)胞。隨機(jī)分為對(duì)照組、單純損傷組、損傷+抑制組,在不同時(shí)間點(diǎn)(損傷后1、6、12、24及48 h)分別應(yīng)用免疫熒光細(xì)胞化學(xué)法測(cè)定磷酸化NF-κB p65蛋白的表達(dá),Annexin V/PI染色法檢測(cè)凋亡、壞死,Western blot檢測(cè)自噬蛋白LC3Ⅱ表達(dá)情況。對(duì)照組為無(wú)劃痕損傷的腦微血管內(nèi)皮細(xì)胞。結(jié)果腦微血管內(nèi)皮細(xì)胞劃痕損傷后NF-κB蛋白表達(dá)于傷后1 h開(kāi)始表達(dá)增強(qiáng),于24 h達(dá)高峰,與對(duì)照組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。凋亡、壞死及自噬各指標(biāo)均隨時(shí)間推移有不同程度升高,與對(duì)照組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。而經(jīng)PDTC處理后上述變化均受不同程度的抑制,與單純損傷組相比差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論劃痕損傷后可激活腦微血管內(nèi)皮細(xì)胞NF-κB表達(dá),同時(shí)可引起腦微血管內(nèi)皮細(xì)胞的自噬、凋亡和壞死。
[Abstract]:Objective to investigate the effect of scratch injury on NF- 魏 B expression and autophagy of cerebral microvascular endothelial cells (MECs) of SD rats in vitro. Methods cultured rat cerebral cortex microvascular endothelial cells in vitro and made scratch injury model. Pyrrolidine dithiocarbamate (PDTCC), an inhibitor of NF- 魏 B, was used to interfere with the scratch injury of cerebral microvascular endothelial cells, and was randomly divided into control group, simple injury group and injury inhibition group. The expression of phosphorylated NF- 魏 B p65 protein was detected by immunofluorescence cytochemistry at different time points (24 and 48 hours after injury).
