【摘要】:目的:探討腫節(jié)風(fēng)注射液與番茄紅素聯(lián)合使用對(duì)ALI大鼠肺損傷炎癥水平的影響。 方法:健康雄性250~280gSD大鼠,50只,均分成5組(隨機(jī)),經(jīng)實(shí)驗(yàn)分組處理后再通過(guò)腹腔用注射器注入6mg/kg的脂多糖建立ALI模型: NC組(n=10):一般對(duì)照組(NS處理); LPS組(模型組,n=10),只給與LPS建模,不給藥處理; LPS+腫節(jié)風(fēng)組(S組,n=10):連續(xù)七天腹腔注射腫節(jié)風(fēng)注射液(2.5g/kg); LPS+番茄紅素組(L組,n=10):連續(xù)2周番茄紅素油樹(shù)脂灌胃(5mg/kg,每天兩次); LPS+腫節(jié)風(fēng)+番茄紅素組(U組,n=10):連續(xù)兩周番茄紅素油樹(shù)脂(5mg/kg,每天兩次)、連續(xù)七天(第2周開(kāi)始)腹腔注射腫節(jié)風(fēng)注射液(2.5g/kg); 分組干預(yù)后第二天建模,建模后2h(T1)、6h(T2)和10h(T3)采集靜脈血,最后一次采血后取大鼠肺組織標(biāo)本,稱重,取右肺中葉,做病理HE切片,分離血清,采用ELISA法檢測(cè)TNF-α和IL-8含量的變化;計(jì)算各組大鼠肺指數(shù),應(yīng)用光學(xué)顯微鏡觀察肺組織病理HE結(jié)果,分級(jí)大鼠肺泡炎并計(jì)分,統(tǒng)計(jì)分析實(shí)驗(yàn)結(jié)果。 結(jié)果: 1.肉眼觀察:NC組大鼠肺臟外觀正常,肺包膜無(wú)腫脹,顏色淡紅,標(biāo)本表面沒(méi)有看到點(diǎn)狀或片狀出血。LPS模型組大鼠肺臟充血比較明顯、標(biāo)本體積腫大,能發(fā)現(xiàn)出血點(diǎn)或出血斑;L組、S組、U組大鼠肺臟標(biāo)本炎癥充血情況好于模型組。 2.肺指數(shù)的比較:LPS組大鼠肺指數(shù)明顯高于其他四個(gè)組(P0.01)其余各干預(yù)組大鼠肺指數(shù)也有不同程度的增高,但均較LPS組增幅;L組、S組、U組與NC組四組之間的肺指數(shù)差異不大。 3.肺組織HE染色比較:NC組大鼠肺組織無(wú)或偶見(jiàn)少量炎癥細(xì)胞浸潤(rùn);LPS組大鼠肺泡之間的間距變得更寬,數(shù)量眾多的各類炎性細(xì)胞浸潤(rùn)在肺間質(zhì),肺泡腔隙被擠壓變窄、有的腔隙內(nèi)可見(jiàn)出血和(或)滲出物;較LPS組,L組、S組和U組炎性滲出情況均有不同程度的減輕,而U組減輕明顯。 4.肺泡炎分級(jí)比較:LPS組肺泡炎評(píng)分均顯著高于其他組;L組、S組和U組肺泡炎評(píng)分小于LPS組(P0.05或P0.01),而大于NC組(P0.05)。L組、S組和U組比較相差不明顯,U組在干預(yù)組(L組、S組、U組)內(nèi)的肺泡炎評(píng)分最低。 5.血清中TNF-α含量變化比較:T1時(shí)間點(diǎn)LPS組大鼠血清中TNF-α含量明顯高于NC組、L組、S組和U組,此差異在統(tǒng)計(jì)學(xué)上有意義(P0.05或P0.01),L組和S組TNF-α含量較NC組均有升高,此差異在統(tǒng)計(jì)學(xué)上有意義(P0.05),U組TNF-α含量較NC組有升高,但無(wú)統(tǒng)計(jì)學(xué)意義,各干預(yù)組之間比較無(wú)統(tǒng)計(jì)學(xué)意義。T2時(shí)間點(diǎn)LPS組大鼠血清中TNF-α含量比NC組、L組、S組和U組仍增高,在有統(tǒng)計(jì)學(xué)上有意義(P0.05),S組和L組TNF-α含量較NC組稍有升高,無(wú)統(tǒng)計(jì)學(xué)意義,U組TNF-α含量較NC組稍有降低,各干預(yù)組之間比較無(wú)統(tǒng)計(jì)學(xué)意義,,T3時(shí)間點(diǎn)LPS組大鼠血清中TNF-α含量仍高于NC組、L組、S組和U組,此差異在統(tǒng)計(jì)學(xué)上有意義(P0.05),但NC組、L組、S組和U組之間比較無(wú)統(tǒng)計(jì)學(xué)意義。隨著實(shí)驗(yàn)時(shí)間延長(zhǎng),LPS組、L組和S組TNF-α含量逐漸降低。 6.血清中細(xì)胞因子IL-8含量變化比較:T1時(shí)間點(diǎn)LPS組大鼠血清中IL-8含量高于NC組、L組、S組和U組,但比較在統(tǒng)計(jì)學(xué)上無(wú)意義,L組、S組和U組IL-8含量大致與NC組持平。T2時(shí)間點(diǎn)LPS組大鼠血清中IL-8含量明顯高于NC組、L組、S組和U組,此差異統(tǒng)計(jì)學(xué)上有意義(P0.05或P0.01),L組IL-8含量比NC組升高,此差異統(tǒng)計(jì)學(xué)上有意義(P0.05),S組和U組TNF-α含量較NC組有升高,此差異未見(jiàn)統(tǒng)計(jì)學(xué)意義,各干預(yù)組之間比較統(tǒng)計(jì)學(xué)上無(wú)意義,T3時(shí)間點(diǎn)LPS組大鼠血清中IL-8含量高于NC組、L組、S組和U組,該差異統(tǒng)計(jì)學(xué)上有意義(P0.05),其余四組之間比較統(tǒng)計(jì)學(xué)上無(wú)意義。隨著實(shí)驗(yàn)時(shí)間延長(zhǎng),LPS組和各干預(yù)組(L組、S組和U組)IL-8含量先升高后降低。 結(jié)論:腫節(jié)風(fēng)注射液聯(lián)合番茄紅素能減輕內(nèi)毒素血癥所致的肺臟炎癥反應(yīng),能在一定程度上對(duì)ALI時(shí)大鼠的受損肺臟予以保護(hù)。
[Abstract]:Objective: To investigate the effect of Zhongjiefeng injection and lycopene combined effect on inflammatory lung injury of ALI rats.
Methods: 50 healthy male 250~280gSD rats were divided into 5 groups (random). After the experimental group was processed, the ALI model was established by injecting 6mg/kg lipopolysaccharide into the abdominal cavity with injector.
Group NC (n=10): general control group (NS treatment);
Group LPS (model group, n=10) was only modeled with LPS, and no drug treatment was given.
LPS+ group (group S, n=10 zjf): seven consecutive days intraperitoneal injection of zjf injection (2.5g/kg);
LPS+ LYC group (group L, n=10) for 2 weeks lycopeneoleoresin gavage (5mg/kg, two times a day);
LPS+ + zjf lycopene group (group U, n=10) for two weeks lycopeneoleoresin (5mg/kg, two times a day), for seven consecutive days (second weeks) intraperitoneal injection of zjf injection (2.5g/kg);
The intervention group second days after modeling, after modeling 2H (T1), 6h (T2) and 10h (T3) in venous blood, blood for the last time after taking the sample, lung tissues of rats weighing, take the right middle lobe, pathological HE slice, separation of serum were measured by ELISA TNF- alpha and IL-8 content the rats in each group; lung index, lung tissue HE results observed by light microscope, grading of alveolitis in rats and scoring, analysis of the experimental results of statistics.
