【摘要】：背景:在皮膚修復(fù)及瘢痕形成過(guò)程中如何控制膠原蛋白的有序形成是值得關(guān)注的熱點(diǎn)。目的:探討堿性成纖維細(xì)胞生長(zhǎng)因子與胰島素樣生長(zhǎng)因子1對(duì)體外培養(yǎng)大鼠骨髓間充質(zhì)干細(xì)胞增殖及膠原合成的影響。方法:體外分離培養(yǎng)大鼠骨髓間充質(zhì)干細(xì)胞,進(jìn)行成脂及成骨分化鑒定,在第3代細(xì)胞培養(yǎng)液中分別添加堿性成纖維細(xì)胞生長(zhǎng)因子、胰島素樣生長(zhǎng)因子1以及兩者聯(lián)合干預(yù),以加入正常培養(yǎng)基為對(duì)照組。培養(yǎng)12,24,48,72,96 h時(shí)采用MTT檢測(cè)細(xì)胞增殖情況,培養(yǎng)10 d時(shí)采用RT-PCR檢測(cè)Ⅰ型膠原、Ⅲ型膠原m RNA表達(dá),Western Blot檢測(cè)Ⅰ型膠原、Ⅲ型膠原蛋白表達(dá)。結(jié)果與結(jié)論:1與對(duì)照組比較,單獨(dú)應(yīng)用堿性成纖維細(xì)胞生長(zhǎng)因子可顯著促進(jìn)骨髓間充質(zhì)干細(xì)胞增殖,抑制Ⅰ型膠原、Ⅲ型膠原表達(dá)。2單獨(dú)應(yīng)用胰島素樣生長(zhǎng)因子1可顯著促進(jìn)骨髓間充質(zhì)干細(xì)胞增殖,促進(jìn)Ⅰ型膠原、Ⅲ型膠原表達(dá)。3兩種生長(zhǎng)因子聯(lián)用相比單獨(dú)應(yīng)用,促進(jìn)增殖效果更為顯著,同時(shí)使Ⅰ型膠原、Ⅲ型膠原表達(dá)趨于正常水平。4結(jié)果表明堿性成纖維細(xì)胞生長(zhǎng)因子、胰島素樣生長(zhǎng)因子1聯(lián)合應(yīng)用在促進(jìn)骨髓間充質(zhì)干細(xì)胞增殖的同時(shí)能夠?qū)Β裥湍z原、Ⅲ型膠原的表達(dá)有所限制,可能有助于控制瘢痕形成及促進(jìn)創(chuàng)傷愈合。
[Abstract]:Background: how to control the orderly formation of collagen in the process of skin repair and scar formation is a hot issue. Objective: to investigate the effect of basic fibroblast growth factor and insulin-like growth factor 1 on in vitro culture. Effects of rat bone marrow mesenchymal stem cells on proliferation and collagen synthesis methods: rat bone marrow mesenchymal stem cells were isolated and cultured in vitro. Adipogenic and osteogenic differentiation were identified. Basic fibroblast growth factor (bFGF), insulin-like growth factor 1 (insulin-like growth factor 1) and combined intervention were added to the third generation cell culture medium, respectively.
