內(nèi)毒素吸附劑SPV對創(chuàng)傷失血性休克模型大鼠腸黏膜損傷的保護(hù)作用

【摘要】:目的:研究內(nèi)毒素吸附劑SPV對創(chuàng)傷失血性休克模型大鼠腸黏膜損傷的保護(hù)作用。方法:將0.5 mg多黏菌素B、5 mg維生素B6、0.3 g蒙脫石散溶于生理鹽水中制成5 ml溶液,即SPV低劑量;中、高劑量藥物成分為低劑量的2、3倍。取大鼠隨機(jī)分為正常對照組、模型組和SPV低、中、高劑量組,每組10只。除正常對照組外,其余各組大鼠通過股動脈插管放血術(shù)復(fù)制創(chuàng)傷失血性休克模型,建模前30 min SPV各劑量組大鼠ig相應(yīng)劑量的SPV。復(fù)蘇后1、4、8、16 h,檢測各組大鼠血清中一氧化氮(NO)、二胺氧化酶(DAO)、內(nèi)毒素及腫瘤壞死因子α(TNF-α)水平,觀察正常對照組、模型組和SPV高劑量組大鼠腸黏膜細(xì)胞特征和損傷指數(shù)。結(jié)果:與正常對照組比較,模型組大鼠腸黏膜細(xì)胞出現(xiàn)壞死的病理現(xiàn)象,血清中NO、DAO、內(nèi)毒素和TNF-α水平均升高(P0.05)。與模型組比較,SPV中、高劑量組大鼠復(fù)蘇后1、4、8、16 h腸黏膜細(xì)胞損傷程度明顯減輕,血清中NO、DAO、內(nèi)毒素和TNF-α水平均降低(P0.05);其余各組比較差異無統(tǒng)計學(xué)意義(P0.05)。結(jié)論:內(nèi)毒素吸附劑SPV能夠降低創(chuàng)傷失血性休克模型大鼠血清中NO、DAO、內(nèi)毒素及TNF-α水平,對創(chuàng)傷失血性休克引起的腸黏膜損傷有保護(hù)作用。
[Abstract]:Objective: to study the protective effect of endotoxin adsorbent SPV on intestinal mucosal injury in rats with traumatic hemorrhagic shock. 0. 3 g montmorillonite powder dissolved in normal saline to make 5 ml solution, that is, low dose of SPV; Rats were randomly divided into normal control group, model group and SPV low, medium and high dose groups, 10 rats in each group, except the normal control group. The model of traumatic hemorrhagic shock was established by femoral artery catheterization and bloodletting in other groups. The rats in each dose group of 30 min SPV were given the corresponding dose of SPV before 30 min before resuscitation. The serum levels of nitric oxide (no), diamine oxidase (diamine oxidase), endotoxin (endotoxin) and tumor necrosis factor 偽 (TNF- 偽) in the serum of rats in each group were measured at 16 h, and the normal control group was observed. Results: compared with the normal control group, the intestinal mucosal cells in the model group and the high dose group of SPV were necrotic. The levels of endotoxin and TNF- 偽 were both increased (P 0.05). Compared with the model group, the injury degree of intestinal mucosal cells in the high dose group was significantly reduced after resuscitation at 1: 4 and 816 h after resuscitation. The levels of NONDAO, endotoxin and TNF- 偽 in serum were all decreased (P 0.05). Conclusion: endotoxin adsorbent SPV can reduce serum NO-DAO in rats with traumatic hemorrhagic shock. The levels of endotoxin and TNF- 偽 have protective effect on intestinal mucosal injury induced by traumatic hemorrhagic shock.
