建立基于超薄切片的高分辨成像技術(shù)解析組織的精細空間結(jié)構(gòu)

【摘要】:目的建立基于超薄切片方法的高分辨三維成像技術(shù),提高Z軸分辨率,并用于組織的精細結(jié)構(gòu)研究。方法組織樣品用LRWhite樹脂滲透聚合后,用鉆石刀將組織切片切成50~1 000 nm的連續(xù)半薄或超薄切片,具體切片的厚度即Z軸的分辨率。切片經(jīng)甲基藍-洋紅染色或多重抗體標記后,利用全自動顯微鏡對連續(xù)切片進行成像,通過二維拼接和三維重構(gòu)獲得組織高分辨的三維空間信息。結(jié)果對人正常結(jié)腸組織和小鼠肝臟進行高分辨成像分析,獲得比傳統(tǒng)顯微鏡更高的分辨效果和更豐富的結(jié)構(gòu)細節(jié)。結(jié)合多重抗體染色方法,對小鼠腦組織微管蛋白的分布進行了高分辨三維成像。結(jié)論所建立的高分辨三維成像技術(shù)適用于各種組織的空間精細結(jié)構(gòu)研究,超薄切片方法同時為超高分辨顯微成像提供Z軸的解決方案。
