【摘要】：目的明確腫瘤壞死因子-α(TNF-α)開放血腫瘤屏障的作用及其可能機(jī)制。方法構(gòu)建體外血腫瘤屏障模型,給予TNF-α后不同時(shí)間點(diǎn),免疫組織化學(xué)法和Western blot法動(dòng)態(tài)檢測(cè)膠質(zhì)瘤細(xì)胞的熱休克蛋白70(heat shock protein70,HSP70)和腺嘌呤核苷受體(adenosine A1receptor,A1R)的表達(dá)水平;熒光分析法檢測(cè)體外血腫瘤屏障模型的通透性改變。結(jié)果給予TNF-α后,膠質(zhì)瘤細(xì)胞內(nèi)HSP70表達(dá)增加,于30 min時(shí)含量最多。C6細(xì)胞內(nèi)A1R的表達(dá)水平也在給予TNF-α后明顯增加,并于120 min達(dá)高峰后逐漸減少。其血腫瘤屏障的通透性亦于給予TNF-α后120 min達(dá)最大后減少。結(jié)論經(jīng)TNF-α作用后的膠質(zhì)瘤細(xì)胞HSP70表達(dá)增加,增加的HSP70可能是通過某種內(nèi)在機(jī)制提高A1R的表達(dá)水平,進(jìn)而增加了血腫瘤屏障的通透性。
[Abstract]:Objective to investigate the role of tumor necrosis factor- 偽 (TNF- 偽) in opening the blood-tumor barrier and its possible mechanism. Methods the blood tumor barrier model was established in vitro. At different time points after TNF- 偽 administration, the heat shock protein 70 (heat shock protein70,HSP70 (HSP70) and adenosine A1 receptor (adenosine A1 receptor) of glioma cells were detected by immunohistochemistry and Western blot assay. A1R) expression level; The permeability of blood tumor barrier model in vitro was detected by fluorescence assay. Results after TNF- 偽 administration, the expression of HSP70 increased in glioma cells, and reached the highest level at 30 min. The expression of A1R in C6 cells also increased significantly after TNF- 偽 administration, and gradually decreased after the peak of 120 min. The permeability of blood tumor barrier was also decreased after TNF- 偽 was given to 120 min. Conclusion the expression of HSP70 in glioma cells treated with TNF- 偽 is increased, and the increased HSP70 may increase the expression of A1R through some internal mechanism, thus increasing the permeability of blood tumor barrier.
