N-of-1無殘留效應(yīng)定量數(shù)據(jù)混合效應(yīng)模型的模擬研究

【摘要】:目的通過對不同時間點(diǎn)間存在相關(guān)、無殘留效應(yīng)的N-of-1定量數(shù)據(jù)進(jìn)行模擬研究,比較不同檢驗(yàn)方法的統(tǒng)計性能。方法模擬參數(shù)設(shè)樣本量為10(模型1),研究周期為3(模型2-4),不同時間點(diǎn)間相關(guān)系數(shù)為0.8(模型5-7),無殘留效應(yīng),根據(jù)固定的效應(yīng)差值產(chǎn)生多元正態(tài)分布數(shù)據(jù),建立配對t檢驗(yàn)、混合效應(yīng)模型和差值的混合效應(yīng)模型。使用效應(yīng)差值估計值的Ⅰ類錯誤、檢驗(yàn)功效、平均誤差(ME),平均絕對誤差(AE),均方誤差(RMSE)評價各種模型。結(jié)果所有模型估計值的均數(shù)都非常接近效應(yīng)差值,所有模型估計值的ME、AE、RMSE都較小。除了模型7,其他模型的Ⅰ類錯誤概率都約等于0.05。隨著效應(yīng)差值的增大,所有模型的檢驗(yàn)功效都隨之增大。當(dāng)兩組的效應(yīng)差值較小時(1.0),模型5的檢驗(yàn)功效最大,模型2至模型4的功效較小。當(dāng)兩組的效應(yīng)差值較大時(≥1.0),所有模型的功效都小于0.010。結(jié)論混合效應(yīng)模型比配對t檢驗(yàn)更適合存在相關(guān)關(guān)系的N-of-1數(shù)據(jù)。混合效應(yīng)模型的效果優(yōu)于差值的混合效應(yīng)模型,效果最優(yōu)的模型是CS結(jié)構(gòu)的混合效應(yīng)模型。
[Abstract]:Aim to compare the statistical performance of different test methods by simulating quantitative N-of-1 data with correlation and no residual effect at different time points. Methods the sample size was 10 (model 1), the research period was 3 (model 2-4), the correlation coefficient between different time points was 0.8 (model 5-7), and there was no residual effect. According to the fixed effect difference, the multivariate normal distribution data are generated, and the paired t test, mixed effect model and mixed effect model of the difference are established. In this paper, we use the class I errors of the estimation of the effect difference to test the effectiveness, the average error (ME), mean absolute error (AE), mean square error (RMSE) to evaluate various models. Results the average values of all models are very close to the effect difference, and the ME,AE,RMSE of all models is smaller. With the exception of model 7, the class I error probability of other models is about 0.05. With the increase of the effect difference, the test effect of all models increases.
