【摘要】：目的復(fù)制尾靜脈注射和腹腔注射接種的裸鼠乳腺癌轉(zhuǎn)移瘤模型,應(yīng)用生物發(fā)光技術(shù)監(jiān)測(cè)腫瘤細(xì)胞在活體內(nèi)的生長(zhǎng)和轉(zhuǎn)移。方法培養(yǎng)轉(zhuǎn)染有熒光素酶基因的人乳腺癌細(xì)胞系MDA-MB-231-luc-D3-h2LN,分別通過(guò)尾靜脈和腹腔接種到裸鼠體內(nèi),每周進(jìn)行生物發(fā)光成像,最后解剖裸鼠體外觀察轉(zhuǎn)移灶。結(jié)果尾靜脈接種模型腫瘤細(xì)胞在體內(nèi)1周時(shí)有明顯減少,之后在肺部定植并迅速增長(zhǎng),于接種后5周體外成像,確認(rèn)肺部轉(zhuǎn)移,其他部位未見(jiàn)腫瘤轉(zhuǎn)移灶;腹腔接種模型的腫瘤細(xì)胞在腹腔形成多個(gè)轉(zhuǎn)移灶,腫瘤細(xì)胞持續(xù)增殖,于接種后6周體外成像,發(fā)現(xiàn)壁層腹膜和臟層腹膜內(nèi)多個(gè)轉(zhuǎn)移灶,各臟器和腹腔以外部位未見(jiàn)腫瘤轉(zhuǎn)移。結(jié)論建立了兩種裸鼠轉(zhuǎn)移瘤模型,并應(yīng)用生物發(fā)光成像技術(shù)實(shí)現(xiàn)了深層部位腫瘤的追蹤監(jiān)測(cè)。
[Abstract]:Objective to establish a nude mouse model of breast cancer metastasis by tail vein injection and intraperitoneal injection, and to monitor the growth and metastasis of tumor cells in vivo by bioluminescence technique. Methods Human breast cancer cell line MDA-MB-231-luc-D3-h2LN, transfected with luciferase gene was inoculated into nude mice via tail vein and intraperitoneal cavity respectively. Bioluminescence imaging was performed every week. Finally, the metastatic foci of nude mice were observed in vitro. Results the tumor cells of caudal vein inoculation model decreased obviously at 1 week in vivo, then were colonized and rapidly increased in lung. After 5 weeks of inoculation, tumor metastasis was confirmed by imaging in vitro, but no tumor metastasis was found in other parts. The tumor cells of the peritoneal inoculation model formed multiple metastatic foci in the abdominal cavity, and the tumor cells proliferated continuously. After 6 weeks of inoculation, imaging in vitro showed that there were multiple metastatic foci in the parietal peritoneum and the visceral peritoneum, and no metastasis was found in all organs and outside the abdominal cavity. Conclusion two models of metastatic tumor in nude mice were established, and bioluminescent imaging was used to monitor the deep tumor.
