腸道菌群失調(diào)促進(jìn)小鼠肺樹突狀細(xì)胞成熟并增強(qiáng)TLR2和TLR4表達(dá)

【摘要】:目的觀察卵清蛋白(OVA)霧化吸入激發(fā)對抗生素所致腸道菌群失調(diào)小鼠肺組織中樹突狀細(xì)胞(DC)成熟度和Toll樣受體2(TLR2)、TLR4表達(dá)的影響。方法將64只3周齡雌性BALB/c小鼠隨機(jī)分為4組,菌群失調(diào)組、菌群失調(diào)加激發(fā)組、激發(fā)組、對照組,每組16只。于實驗第14天用流式細(xì)胞術(shù)檢測肺組織中DC成熟度(MHC-Ⅱ類分子)及其膜表面TLR2、TLR4的表達(dá)水平,采用免疫組織化學(xué)法檢測肺組織中轉(zhuǎn)化生長因子β1(TGF-β1)、白細(xì)胞介素-6(IL-6)蛋白表達(dá),ELISA檢測支氣管肺泡灌洗液(BALF)中TGF-β1、IL-6的產(chǎn)生水平。結(jié)果與對照組和激發(fā)組比較,菌群失調(diào)組及菌群失調(diào)加激發(fā)組小鼠肺組織中DC細(xì)胞膜表面MHC-Ⅱ類分子及TLR2、TLR4的表達(dá)均增加,但這些分子在上述2組中的表達(dá)無差異(P0.05)。與其他3組比較,菌群失調(diào)加激發(fā)組小鼠肺組織及BALF中TGF-β1、IL-6水平增高。結(jié)論腸道菌群失調(diào)使肺部DC成熟度增高,TLR2、TLR4表達(dá)增強(qiáng)。
