【摘要】：研究背景隨著社會(huì)的進(jìn)步、發(fā)展和不斷變革,人們?cè)谙硎苤@一切帶來(lái)的高效、便捷生活方式的同時(shí),也承受著越來(lái)越大的壓力。尤其是生活在城市的居民,要面臨更多的工作、學(xué)習(xí)、擇業(yè)等壓力,面對(duì)更激烈的競(jìng)爭(zhēng),這時(shí),人們往往更容易處于各種急、慢性應(yīng)激狀態(tài)。適度的應(yīng)激可以提高機(jī)體的適應(yīng)能力,使人保持上進(jìn)心,但是過(guò)度的應(yīng)激持續(xù)時(shí)間長(zhǎng),就會(huì)成為適應(yīng)性的負(fù)擔(dān),久而久之就會(huì)導(dǎo)致機(jī)體焦慮。研究表明,長(zhǎng)期處于過(guò)度應(yīng)激狀態(tài)易于導(dǎo)致焦慮,目前80%的疾病均與過(guò)度應(yīng)激有密切的關(guān)系。長(zhǎng)時(shí)間的持續(xù)應(yīng)激,神經(jīng)內(nèi)分泌系統(tǒng)紊亂,引起相關(guān)炎性細(xì)胞因子的分泌、合成和轉(zhuǎn)輸異常。熱休克蛋白70(HSP70)作為重要的保護(hù)蛋白,參與調(diào)控炎性細(xì)胞因子的表達(dá)。臨床上的抗焦慮藥物存在一定的副作用,研究者逐步把目光投向了中醫(yī)學(xué)領(lǐng)域。研究目的本課題主要探討逍遙散對(duì)慢性束縛應(yīng)激焦慮模型大鼠胃黏膜HSP70及相關(guān)炎性細(xì)胞因子的調(diào)節(jié)作用,以亟探索逍遙散保護(hù)因情志不適而導(dǎo)致的胃黏膜損傷的作用機(jī)制,為臨床應(yīng)用逍遙散治療因情志不適所導(dǎo)致的胃黏膜損傷提供實(shí)驗(yàn)依據(jù)。研究方法將48只健康的雄性SD大鼠(體重為220-250g)隨機(jī)分為正常組、模型組、逍遙散組、咯利普蘭組,每組分別12只。分組后的大鼠適應(yīng)性喂養(yǎng)10d,實(shí)驗(yàn)組均以慢性束縛應(yīng)激的方法制備焦慮大鼠模型:每天3h,連續(xù)14d。逍遙散組灌胃逍遙散懸浮液,而正常組和模型組分別灌胃去離子水,咯利普蘭組注射相應(yīng)藥物。分別在第0、7、14d對(duì)大鼠進(jìn)行體重測(cè)量、高架十字迷宮行為學(xué)測(cè)試;在實(shí)驗(yàn)的第0、14d做新環(huán)境進(jìn)食抑制實(shí)驗(yàn)。每天定時(shí)觀察大鼠的精神狀態(tài)、身體姿勢(shì)、皮毛色澤、活動(dòng)度、對(duì)束縛的反應(yīng)程度、眼黏膜色澤、耳廓色澤以及糞便等一般情況。通過(guò)免疫組化法檢測(cè)胃黏膜HSP70的表達(dá)量。通過(guò)ELISA實(shí)驗(yàn)檢測(cè)血清中IL-6、IL-10等相關(guān)炎性細(xì)胞因子的含量。結(jié)果1.一般狀態(tài),兩周造模結(jié)束時(shí),正常組、逍遙散組和咯利普蘭組大鼠明顯比模型組大鼠動(dòng)作靈活,叫聲柔和,嘶叫、反抗、掙扎的次數(shù)也明顯較少,精神狀態(tài)也尚可,毛發(fā)也比模型組整齊,糞便干濕適中呈球狀;而模型組大鼠萎靡不振,毛發(fā)凌亂,色澤不明,蜷伏在角落里呈警惕狀態(tài),束縛時(shí)的反抗、嘶叫、掙扎也比一周前稍有減弱。2.體重變化,造模前1d及造模后第7d,各組大鼠體重未見(jiàn)明顯差異,到第14d,與模型組相比,其余三組大鼠平均體重高于模型組,有統(tǒng)計(jì)學(xué)差異(P0.05),與正常組相比,逍遙散組及咯利普蘭組大鼠體重?zé)o顯著性差異。3.新環(huán)境進(jìn)食抑制實(shí)驗(yàn),在實(shí)驗(yàn)開(kāi)始時(shí),各組無(wú)統(tǒng)計(jì)學(xué)差異,而造模后的第14d,模型組時(shí)間明顯多于逍遙散組。4.高架十字迷宮實(shí)驗(yàn),兩周造模實(shí)驗(yàn)結(jié)束后,與正常組相比,模型組大鼠封閉臂進(jìn)入次數(shù)減少,停留時(shí)間延長(zhǎng);與正常組相比,其余三組雙側(cè)開(kāi)放臂進(jìn)入次數(shù)減少且停留時(shí)間減少,有統(tǒng)計(jì)學(xué)意義;與模型組相比,逍遙散組大鼠封閉臂進(jìn)入次數(shù)增多且停留時(shí)間短,但均無(wú)統(tǒng)計(jì)學(xué)差異。5.免疫組化實(shí)驗(yàn),模型組大鼠胃黏膜細(xì)胞表達(dá)的HSP70明顯低于正常組,具有統(tǒng)計(jì)學(xué)意義(P0.05),與模型組大鼠相比,逍遙散組大鼠陽(yáng)性表達(dá)量明顯升高,且具有統(tǒng)計(jì)學(xué)意義(P0.01);逍遙散組和咯利普蘭組間無(wú)統(tǒng)計(jì)學(xué)差異,逍遙散組表達(dá)量略高于咯利普蘭組。6.ELISA實(shí)驗(yàn),模型組大鼠血清中的IL-1β、IL-6、IL-10和TNF-α含量明顯上升,高于其它三組,逍遙散組和咯利普蘭組間無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論14d慢性束縛應(yīng)激可以導(dǎo)致大鼠焦慮,逍遙散可通過(guò)促進(jìn)胃黏膜產(chǎn)生大量HSP70的方式來(lái)抑制相關(guān)炎性細(xì)胞因子的釋放,進(jìn)而起到保護(hù)胃黏膜的作用,為臨床應(yīng)用逍遙散治療因情志不適所導(dǎo)致的胃黏膜損傷提供實(shí)驗(yàn)依據(jù)。
[Abstract]:With the progress, development and continuous change of the society, people are enjoying more and more pressure while enjoying the high efficiency and convenient way of living. Moderate stress can improve the adaptability of the body, keep the person into the heart, but excessive stress lasts long, it will become the burden of adaptability, and it will lead to the anxiety of the body for a long time. The study shows that the excessive stress is easy to cause anxiety for a long time, and 80% of the diseases are all over the time. Degree stress has a close relationship. Long lasting stress, neuroendocrine system disorder, secretion of related inflammatory cytokines, synthesis and transtransmission abnormalities. Heat shock protein 70 (HSP70) is an important protective protein and participates in the regulation of the expression of inflammatory cytokines. There are certain side effects in clinical anti anxiety drugs. The purpose of this study is to explore the effect of Xiaoyao Powder on the gastric mucosal HSP70 and related inflammatory cytokines in rats with chronic restraint stress anxiety, in order to explore the mechanism of the action of Xiaoyao Powder to protect the gastric mucosa caused by emotional discomfort. 48 healthy male SD rats (220-250g) were randomly divided into the normal group, the model group, the Xiaoyao San group, the rrrl group, and the group of 12.
