解剖部位差異對(duì)脂肪干細(xì)胞生物學(xué)行為及成骨分化能力的影響

【摘要】:目的探討不同解剖部位來(lái)源的脂肪干細(xì)胞在增殖、活性、成骨分化能力上的差異。方法實(shí)驗(yàn)所用脂肪均取自3只雌性新西蘭大白兔脂肪組織,按解剖部位分組,分別為:腹股溝組、頸背部組、腎周組。各組脂肪經(jīng)體外分離獲得脂肪干細(xì)胞,并對(duì)其細(xì)胞增殖、活性、成骨分化能力進(jìn)行檢測(cè)。結(jié)果在MTT細(xì)胞增殖實(shí)驗(yàn)中,腹股溝組和頸背部組細(xì)胞增殖能力均明顯優(yōu)于腎周組(P0.05),而腹股溝組和頸背部組間無(wú)差異(P0.05);在活/死細(xì)胞染色檢測(cè)細(xì)胞活性實(shí)驗(yàn)中,3組間細(xì)胞活性差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);在半定量分析堿性磷酸酶(ALP)活性及qRT-PCR檢測(cè)Runx2、COLL-1、OC表達(dá)以反映成骨分化能力的實(shí)驗(yàn)中,來(lái)源于皮下脂肪的兩組(腹股溝組和頸背部組)的ALP活性高于腎周組(P0.05),并且腹股溝組比頸背部組的成骨能力更強(qiáng)(P0.05)。結(jié)論來(lái)源于皮下脂肪的脂肪干細(xì)胞具有更好的增殖和分化能力,其中腹股溝皮下脂肪來(lái)源的脂肪干細(xì)胞的性能更加優(yōu)越。
[Abstract]:Objective to investigate the difference of proliferation, activity and osteogenic differentiation of adipose stem cells from different anatomical sites.Methods the adipose tissue of 3 female New Zealand rabbits were divided into three groups: inguinal group, cervical dorsal group and perirenal group.Adipose stem cells were isolated from adipose cells in vitro, and the proliferation, activity and osteogenic differentiation of adipose stem cells were measured.Results in MTT cell proliferation assay,The proliferative ability of groin group and cervical dorsal group were significantly better than that of perirenal group, but there was no difference between inguinal group and cervical back group.In the experiment of semi-quantitative analysis of alkaline phosphatase (ALP) activity and detection of Runx2COLL-1OC expression by qRT-PCR to reflect the osteogenic differentiation ability,Conclusion the adipose stem cells derived from subcutaneous adipose cells have better proliferation and differentiation ability, and the adipose stem cells derived from inguinal subcutaneous adipose cells have better performance.
