四逆湯對同型半胱氨酸損傷的EAhy926細胞caveolin-1和eNOS表達的影響

【摘要】:目的:探討四逆湯(Sini decoction,SND)防治內(nèi)皮細胞損傷的機制及陷窩蛋白1(caveolin-1)和一氧化氮(NO)系統(tǒng)在其中的作用。方法:建立同型半胱氨酸(Hcy)損傷的人臍靜脈內(nèi)皮融合細胞(EAhy926細胞)模型,觀察四逆湯的保護作用及其對NO系統(tǒng)和caveolin-1的影響。結(jié)果:Hcy加入后細胞生長緩慢,貼壁細胞數(shù)明顯減少,NO濃度明顯減低(P0.05),caveolin-1 mRNA和蛋白表達明顯增強(P0.05),內(nèi)皮型NO合酶(eNOS)mRNA和蛋白表達明顯減弱(P0.05);四逆湯處理組貼壁細胞及細胞形態(tài)明顯改善,NO濃度較Hcy模型組明顯升高(P0.05),caveolin-1 mRNA和蛋白表達較Hcy模型組明顯減弱(P0.05),eNOS mRNA和蛋白表達較Hcy模型組明顯增強(P0.05),其中以四逆湯1.0 kg/L+Hcy 4.0μmol/L組最明顯。結(jié)論:Hcy可能通過增加caveolin-1表達和抑制eNOS表達而損傷人臍靜脈內(nèi)皮細胞,而四逆湯通過抑制caveolin-1表達和增加eNOS的表達拮抗Hcy對細胞的損傷。
[Abstract]:Objective: to investigate the mechanism of Sini decoctionn SND in the prevention and treatment of endothelial cell injury and the role of caveolin-1) and nitric oxide (no) system in the prevention and treatment of endothelial injury. Methods: a model of human umbilical vein endothelial fusion cell line EAhy926 induced by Hcyine homocysteine injury was established. To observe the protective effect of Sini decoction and its effect on no system and caveolin-1. The number of adherent cells significantly decreased the concentration of no decreased significantly, the expression of mRNA and protein increased significantly, the expression of endothelium-type no synthase (ENOSN) mRNA and protein decreased significantly (P0.05), and the adherent cells and cell morphology of Sini decoction group significantly improved the concentration of no compared with Hcy model. The expression of mRNA and protein was significantly increased in group A than that in group Hcy. Conclusion the expression of P0.05Enos mRNA and protein in group A is significantly lower than that in group Hcy, especially in the group of 1.0 kg/L Hcy 4.0 渭 mol/L with Sini decoction. Conclusion it may be possible to increase the expression of caveolin-1 and inhibit the expression of eNOS in Sini decoction (1.0 kg/L Hcy 4.0 渭 mol/L). Expression and damage human umbilical vein endothelial cells, Sini decoction antagonized the damage of Hcy by inhibiting the expression of caveolin-1 and increasing the expression of eNOS.
