TAK1抑制TGF-β1誘導(dǎo)小鼠巨噬細(xì)胞RAW264.7凋亡的實(shí)驗(yàn)研究

【摘要】:目的探討TAK1對(duì)TGF-β1誘導(dǎo)小鼠巨噬細(xì)胞凋亡的影響。方法電穿孔法將TAK1等目的基因質(zhì)粒瞬時(shí)轉(zhuǎn)染小鼠巨噬細(xì)胞RAW264.7,pEGFP-CⅡ觀察轉(zhuǎn)染效率,采用Real time-PCR及Western blot檢測(cè)RAW264.7細(xì)胞轉(zhuǎn)染后TAK1基因和蛋白的表達(dá)水平,流式細(xì)胞術(shù)檢測(cè)過(guò)表達(dá)TAK1后對(duì)RAW264.7細(xì)胞凋亡的影響。結(jié)果 TAK1轉(zhuǎn)染組、TAK1+TAB1共轉(zhuǎn)染組細(xì)胞凋亡率明顯低于空質(zhì)粒對(duì)照組。結(jié)論巨噬細(xì)胞中過(guò)表達(dá)TAK1能抑制TGF-β1刺激所誘導(dǎo)的凋亡,且共轉(zhuǎn)染TAB1+TAK1組抑制作用最明顯。
