【摘要】：目的本研究擬通過(guò)白黎蘆醇對(duì)IBS動(dòng)物模型進(jìn)行治療,以觀察動(dòng)物焦慮、抑郁等肝郁情志表現(xiàn)是否能得到改善,并通過(guò)腸道動(dòng)力和內(nèi)臟敏感性檢測(cè),以觀察白藜蘆醇是否有可能成為治療IBS的潛在安全有效的藥物。方法1.建立慢急性聯(lián)合應(yīng)激腸易激綜合征模型ICR小鼠,小鼠隨機(jī)分成8組,每組12只。正常對(duì)照組、慢急性聯(lián)合應(yīng)激模型組、白藜蘆醇作用組(分低、中、高三個(gè)不同劑量,分別為2.5、5、10 mg/kg,灌胃)及三組陽(yáng)性藥物對(duì)照組。通過(guò)糖水消耗實(shí)驗(yàn)、曠場(chǎng)實(shí)驗(yàn)、大理石掩埋實(shí)驗(yàn)和高架十字迷宮實(shí)驗(yàn)等檢測(cè)小鼠抑郁、焦慮樣行為。并檢測(cè)腹壁撤退反射檢測(cè)內(nèi)臟敏感性、色素推進(jìn)實(shí)驗(yàn)檢測(cè)腸道動(dòng)力。結(jié)果慢急性聯(lián)合應(yīng)激腸易激綜合征小鼠模型隨著時(shí)間的延長(zhǎng)表現(xiàn)出顯著的抑郁,高中低三個(gè)劑量的白藜蘆醇均能改善其抑郁行為。IBS模型小鼠均表現(xiàn)出恐高的焦慮,高劑量組白藜蘆醇可以減輕其焦慮行為。在大理石掩埋實(shí)驗(yàn)中,IBS小鼠模型均表現(xiàn)出焦慮,高中低三個(gè)劑量的白藜蘆醇均可改善其焦慮癥狀。而在內(nèi)臟敏感性方面,在腸道給予稍大量空氣后顯著升高,高中低三個(gè)劑量的白黎蘆醇均可降低IBS模型小鼠的腸道高敏感性。在腸道動(dòng)力方面,IBS模型小鼠表現(xiàn)出腸道動(dòng)力的減弱,中高劑量白藜蘆醇顯著逆轉(zhuǎn)腸道動(dòng)力的減弱。說(shuō)明IBS小鼠存在抑郁焦慮等情志變化,并伴有腸道高敏感性和腸道動(dòng)力減慢,而白藜蘆醇可以逆轉(zhuǎn)上述這些變化。本研究為白藜蘆醇作為腦腸雙靶點(diǎn)作用的治療IBS的安全有效的藥物提供了實(shí)驗(yàn)依據(jù)。結(jié)論慢急性聯(lián)合應(yīng)激腸易激綜合征模型小鼠存在焦慮、抑郁等情志變化,并有腸道敏感性增加、腸道動(dòng)力減慢等表現(xiàn);白藜蘆醇能夠逆轉(zhuǎn)腸易激綜合征小鼠焦慮抑郁等癥狀,并改善腸道高敏感和腸道動(dòng)力變化。
[Abstract]:Objective to observe whether the expression of depression and anxiety in IBS animal model can be improved, and to detect intestinal motility and visceral sensitivity. To observe whether resveratrol might be a potentially safe and effective drug in the treatment of IBS. Methods 1. Establish a model of chronic acute stress irritable bowel syndrome (ICR) mice. The mice were randomly divided into 8 groups with 12 rats in each group. Chronic acute combined stress model group, resveratrol group (low, middle and high three different doses of 2.5 渭 g / kg, intragastric perfusion) and three groups of positive drug control group. Through sugar consumption test, open field test, Marble burying test and elevated cross maze test were used to detect depression and anxiety in mice, and abdominal wall retreating reflex to detect visceral sensitivity. Results the mice model of irritable bowel syndrome with chronic acute stress showed significant depression over time. Three doses of resveratrol could improve the depressive behavior. The IBS model mice showed the anxiety of fear of heights, the high dose group of resveratrol alleviated their anxiety behavior, and the mice model of IBS showed anxiety in marble burying experiment. High and low doses of resveratrol improved anxiety symptoms. In visceral sensitivity, it increased significantly after the gut was given a little more air. Three doses of high and low doses of silylitol could reduce the high sensitivity of intestinal tract in IBS model mice, and the intestinal motility of IBS model mice was decreased in terms of intestinal motility. Resveratrol significantly reversed the decrease of intestinal motility in IBS mice, indicating that depression, anxiety and other emotional changes were associated with intestinal hypersensitivity and decreased intestinal motility in IBS mice. Resveratrol can reverse these changes. This study provides an experimental basis for the safety and efficacy of resveratrol in the treatment of IBS. Conclusion there is anxiety in the model mice of irritable bowel syndrome induced by chronic acute stress. Resveratrol could reverse symptoms such as anxiety and depression in irritable bowel syndrome mice and improve intestinal hypersensitivity and intestinal motility.
