良附丸對實(shí)驗(yàn)性胃潰瘍模型大鼠的藥效學(xué)研究

【摘要】:目的:本實(shí)驗(yàn)建立四種實(shí)驗(yàn)性胃潰瘍動(dòng)物模型,觀察良附丸對模型動(dòng)物的防治作用,并明確其防治效果;進(jìn)而觀察良附丸各劑量組對實(shí)驗(yàn)性胃潰瘍的藥效,并初步探討良附丸抗實(shí)驗(yàn)性胃潰瘍的作用機(jī)制。方法:實(shí)驗(yàn)分兩個(gè)階段進(jìn)行,第一階段明確良附丸對四種實(shí)驗(yàn)性胃潰瘍的防治效果及良附丸各劑量組對實(shí)驗(yàn)性胃潰瘍的藥效。四種實(shí)驗(yàn)性胃潰瘍模型分別為幽門結(jié)扎型、束縛-水浸應(yīng)激型、消炎痛型及醋酸涂抹型。第一步將每種模型分為3組:良附丸組、模型組、陽性對照組,各組大鼠分別進(jìn)行實(shí)驗(yàn)、取材,觀察胃組織病理變化,測量潰瘍指數(shù)并計(jì)算潰瘍愈合率;根據(jù)第一步實(shí)驗(yàn)所得結(jié)果,第二步探討良附丸各劑量抗束縛-水浸應(yīng)激型胃潰瘍的藥效,分為6組,良附丸高、中、低劑量組,模型組,空白對照組,陽性對照組,實(shí)驗(yàn)步驟同第一步。根據(jù)第一階段所得實(shí)驗(yàn)結(jié)果,第二階段探討良附丸對束縛-水浸應(yīng)激型胃潰瘍的作用機(jī)制,分為4組:空白組、良附丸組、模型組、陽性對照組,預(yù)防性給藥4天后造模,腹主動(dòng)脈取血、取下丘腦、取胃,觀察胃組織損傷情況,測量潰瘍指數(shù)計(jì)算潰瘍愈合率,測定大鼠血清中SOD、MDA、NO和腦組織中5-HT的含量。結(jié)果:良附丸對幽門結(jié)扎型和束縛-水浸應(yīng)激型胃潰瘍均有防治作用,能使胃黏膜損傷程度降低,潰瘍抑制率升高,且對束縛-水浸應(yīng)激型胃潰瘍抑制率高于幽門結(jié)扎型胃潰瘍。良附丸高劑量組及陽性對照組可顯著降低潰瘍指數(shù)(p0.01),且效果優(yōu)于中、低劑量組。良附丸高劑量組較模型組相比,能夠升高大鼠血清中NO、SOD,降低血清中MDA及腦組織中5-HT含量(p0.05,p0.01)。結(jié)論:1.良附丸對幽門結(jié)扎型和束縛-水浸應(yīng)激型胃潰瘍均有防治作用。2.束縛-水浸應(yīng)激型胃潰瘍:良附丸高、中劑量組對束縛-水浸應(yīng)激型胃潰瘍均有防治作用,且高劑量組的防治效果優(yōu)于其他劑量組。3.良附丸促束縛-水浸應(yīng)激型大鼠胃潰瘍愈合的作用機(jī)制可能通過升高大鼠血清中保護(hù)因子SOD、NO的含量,降低血清MDA及腦組織5-HT含量來實(shí)現(xiàn)的。
[Abstract]:Objective: to establish four animal models of experimental gastric ulcer and observe the preventive and therapeutic effects of Liangfu pills on model animals. Then observe the effect of Liangfu Pill on experimental gastric ulcer and explore the mechanism of Liangfu Pill on experimental gastric ulcer. Methods: the experiment was carried out in two stages. In the first stage, the preventive and therapeutic effects of Liangfu Pill on four kinds of experimental gastric ulcer were clarified. The four experimental gastric ulcer models were pyloric ligation type and irreducible water-immersion stress type. In the first step, each model was divided into three groups: Liangfu pill group, model group, positive control group, each group of rats were experimented, taken materials, observed the pathological changes of gastric tissue. Ulcer index was measured and ulcer healing rate was calculated. According to the results of the first step, the second step was to investigate the efficacy of Liangfu Pill in anti-restraint and water-immersion stress type gastric ulcer. It was divided into 6 groups: high, middle and low dose group, model group and blank control group. According to the experimental results obtained in the first stage, the mechanism of Liangfu Pill on irreducible water-immersion stress gastric ulcer was studied in the second stage, and was divided into four groups: blank group and Liangfu Pill group. Model group, positive control group, preventive administration 4 days after the model, abdominal aorta blood, hypothalamus, stomach, observe gastric tissue injury, ulcer index to calculate ulcer healing rate. The content of 5-HT in brain tissue and serum SOD MDADAN were determined. Results: Liangfu pills could prevent and treat gastric ulcer of pyloric ligation type and irreducible water-immersion stress type. It can reduce the degree of gastric mucosal injury and increase the inhibition rate of ulcer. The inhibition rate of gastric ulcer in the restraint water immersion stress type was higher than that in the pyloric ligation type. The high dose group of Liangfu pills and the positive control group could significantly reduce the ulcer index (P 0.01), and the effect was better than that in the middle group. Compared with the model group, the high dose group of Liangfu pills could increase the serum NON-SOD, decrease the MDA in serum and 5-HT content in brain tissue. Conclusion: Liangfu Pill can prevent and cure gastric ulcer of pyloric ligation type and bound water stress type. 2. Restraint and water immersion stress type gastric ulcer: Liangfu Pill is high. The middle dose group had the preventive and therapeutic effect on the gastric ulcer induced by irreducible water immersion stress. The preventive and therapeutic effect of high dose group was better than that of other dosage group. 3. The mechanism of Liangfu pills promoting the healing of gastric ulcer induced by water immersion stress might be by increasing the content of protective factor SODN in the serum of rats. To reduce serum MDA and brain tissue 5-HT content to achieve.
