Suppression subtractive hybridization and real time RT-PCR reveal differentially expressed immune-related genes in crucian carp Carassius auratus gibelio challenged with Cyprinid herpesvirus 2

【摘要】：Since 2011,Cyprinid herpesvirus 2(CyHV-2)has been associated with the mortality ofcultured crucian carp Carassius auratus gibelio in east China,which was never reported beforedespite the fact that experimental infection of crucian carp by CyHV-2 had been successful inlaboratories outside of China.In this study,virus-free crucian carps were infected with 108CyHV-2 per fish by injection method,which resulted in a death rate of 95%with typical symptom.To study the role of innate immunity in response to viral infection,virus-induced genes wereidentified using suppression subtractive hybridisation(SSH)from moribund crucian carpchallenged with CyHV-2.A total of 500 clones from SSH forward library were sequenced,50genes were characterized to play a possible role in antiviral process.Among them,16 genes wereCyHV-2-encoded viral genes and 8 genes were immune-related host genes according to theirGene Ontology annotations of biological processes and molecular functions.The comparison ofinfected to uninfected crucian carps were performed using a quantitative real time RT-PCRapproach,which showed corresponding expression profiles for 7 of the 8 identifiedimmune-related genes including nuclear factor of kappa light polypeptide gene enhancer in B-cellsinhibitor(nfkbiab),MHC class I(MHC I),intelectin,interleukin-11(IL-11),interleukin-10receptor(IL-10R),complement component C7-1(C7-1),purine nucleoside phosphorylase 5a(PNP5a)and serum amyloid A protein(SAA).Systematic tissue distribution of the identifiedimmune-related genes correlated with their potential involvement in immune response to CyHV-2,which displayed wide tissue tropism.These findings thus provide a basis for characterizing theantiviral immune-response of crucian carps and might be employed to select marker genes ofinterest in breeding crucian carps with CyHV-2 resistance.
