【摘要】：為了研究干擾素誘導(dǎo)的跨膜蛋白3(Interferon-induced transmembrane protein 3,IFITM 3)rs12252單核苷酸多態(tài)性(Single nucleotide polymorphism,SNP)與乙型流感臨床嚴(yán)重程度的相關(guān)性,在吉林省和湖南省收集了181名乙型流感輕癥病例和83名乙型流感重癥病例全血標(biāo)本,提取兩組病例基因組DNA并擴(kuò)增相應(yīng)的目的片段后,采用Sanger測(cè)序的方法對(duì)ITIM3rs12252SNP進(jìn)行基因分型。同時(shí)選用千人基因組103名中國(guó)北京地區(qū)漢族人(CHB)為一般人群對(duì)照。統(tǒng)計(jì)分析結(jié)果表明,輕癥病例IFITM 3rs12252基因型頻率分布與一般人群對(duì)照無(wú)顯著性差異,但重癥病例rs12252CC基因型頻率顯著高于一般人群對(duì)照組。乙型流感重癥病例IFITM3rs12252C等位基因頻率顯著高于輕癥病例(P0.05),攜帶rs12252-C等位基因發(fā)展為乙型流感重癥的風(fēng)險(xiǎn)是rs12252-T等位基因的1.818倍(Odds Ratio=1.818,95%CI:1.241~2.664)。并且重癥病例中rs12252CC基因型頻率顯著高于輕癥病例(P0.05),表明rs12252CC基因型與乙型流感重癥相關(guān)。
[Abstract]:In order to study the correlation between the polymorphism of the interferon-induced transmembrane protein 3 (IFITM 3) rs12252 and the clinical severity of influenza B, In Jilin province and Hunan province,181 cases of influenza B and 83 of the whole blood of the severe case of influenza B were collected. The genomic DNA of the two groups was extracted and the corresponding target fragment was amplified, and the gene of the ITIM3rs12252SNP was genotyped by using the method of Sanger sequencing. At the same time,103 Chinese Han people (CHB) were selected as the control of the general population. The results of the statistical analysis showed that the frequency distribution of the IITM 3 rs12252 genotype was not significantly different from that of the control group in the general population, but the frequency of rs12252CC in the severe case was significantly higher than that of the control group in the general population. The frequency of the IFITMrs12252C allele in the severe case of influenza B was significantly higher than that of the mild case (P0.05). The risk of the development of the rs12252-C allele was 1.818 times that of the rs12252-T allele (Ods Ratio = 1.818,95% CI: 1.241-2.664). The genotype frequency of rs12252CC was significantly higher in the severe cases (P0.05), indicating that the rs12252CC genotype was related to the severe type of influenza B.
