【摘要】：目的：探討泡球蚴囊液對(duì)體外大鼠肝星狀細(xì)胞（HSC-T6）增殖和凋亡影響的初步研究。 方法： HSC-T6細(xì)胞進(jìn)行體外培養(yǎng)，用CCK-8檢測(cè)并繪制HSC-T6的生長(zhǎng)曲線(xiàn)、不同濃度泡球蚴囊液（0mg/ml-13.5mg/ml）干預(yù)24h時(shí)的增殖活性，并應(yīng)用熒光倒置顯微鏡觀察囊液作用前后的HSC-T6形態(tài)變化；采用流式細(xì)胞儀檢測(cè)并分析HSC-T6細(xì)胞周期和凋亡情況。 結(jié)果：①不同濃度泡球蚴囊液（0mg/ml-13.5mg/ml）干預(yù)的24h后部分細(xì)胞固縮，，呈脫落前兆，而部分HSC-T6細(xì)胞收縮成長(zhǎng)梭形，細(xì)胞生出許多細(xì)長(zhǎng)的偽足。②濃度為0mg/ml-0.85mg/ml時(shí)則對(duì)HSC-T6細(xì)胞增殖活性有促進(jìn)作用，濃度為0.85mg/ml-13.5mg/ml時(shí)則對(duì)HSC-T6細(xì)胞增殖活性有明顯抑制作用，且都有時(shí)間和濃度依懶性。③流式細(xì)胞儀結(jié)果顯示隨著泡球蚴囊液濃度（0mg/ml-6.8mg/ml）的增加，大鼠HSC-T6細(xì)胞的凋亡率逐漸增加。 結(jié)論：在一定濃度范圍內(nèi)，泡球蚴囊液對(duì)體外培養(yǎng)大鼠肝星狀細(xì)胞增殖有一定的促進(jìn)作用，并可促進(jìn)其凋亡；泡球蚴囊液能夠促進(jìn)肝星狀細(xì)胞由G0/G1期進(jìn)入S期。
[Abstract]:Aim: to investigate the effect of alveolar hydatid cyst fluid on proliferation and apoptosis of rat hepatic stellate cells (HSC-T6) in vitro. Methods: HSC-T6 cells were cultured in vitro, the growth curve of HSC-T6 was detected and drawn by CCK-8, the proliferation activity of different concentrations of hydatid cyst fluid (0mg/ml-13.5mg/ml) for 24 hours was observed, the morphological changes of HSC-T6 were observed by fluorescence inverted microscope, and the cell cycle and apoptosis of HSC-T6 cells were detected and analyzed by flow cytometry. Results: 1 after 24 hours of intervention with different concentrations of alveolar hydatid cyst fluid (0mg/ml-13.5mg/ml), some cells shrank and showed exfoliative precursors, while some HSC-T6 cells contracted and grew fusiform, and the cells produced many slender pseudopods. 2 the concentration of 0mg/ml-0.85mg/ml promoted the proliferation activity of HSC-T6 cells. At the concentration of 0.85mg/ml-13.5mg/ml, the proliferation activity of HSC-T6 cells was significantly inhibited, and both time and concentration were lazy. 3 the results of flow cytometry showed that the apoptosis rate of rat HSC-T6 cells increased gradually with the increase of the concentration of alveolar hydatid cyst fluid (0mg/ml-6.8mg/ml). Conclusion: in a certain concentration range, the alveolar hydatid cyst fluid can promote the proliferation and apoptosis of rat hepatic stellate cells in vitro, and the alveolar hydatid cyst fluid can promote the hepatic stellate cells from G0/G1 phase to S phase.
