2015年佛山市首例本地登革熱病例及蚊媒的病原分子溯源

【摘要】:目的對2015年佛山市報告的首例本地登革熱疑似病例進行分子流行病學調(diào)查,確定病毒的基因型及感染來源。方法采集疑似患者血清標本、疫點白紋伊蚊成蚊及孑孓,采用Real-time PCR進行登革病毒基因檢測及分型;用C6/36細胞分離登革病毒,采用套式RT-PCR擴增病毒E蛋白基因,進行序列測定和同源性比較分析。結果從患者血清和伊蚊研磨液中分別檢測到1型登革病毒核酸,兩者的病毒E蛋白基因核苷酸序列同源性為100%。從患者血清的C6/36細胞培養(yǎng)中觀察到登革病毒特異的CPE。將分離株登革病毒命名為FS-D15001。通過對不同地區(qū)的登革毒株進行E蛋白基因核苷酸序列同源性比較,顯示FS-D15001與2014年同一地區(qū)登革熱患者分離毒株的同源性較高,為99.9%。結論經(jīng)分子溯源證實2015年佛山市首例本地登革熱病例由I型登革病毒引起,通過白紋伊蚊傳播,該毒株在當?shù)亓餍锌赡芤殉掷m(xù)數(shù)年。
