利福平對(duì)結(jié)核分枝桿菌在不同植入材料界面黏附力及生物膜形成的影響

【摘要】:目的通過(guò)體外實(shí)驗(yàn)研究利福平對(duì)結(jié)核分枝桿菌(MTB)在不同植入材料界面黏附力及生物膜形成的影響。方法隨機(jī)選取鈷鉻鉬合金、鈦合金、聚乙烯樣品各兩塊分別置入3支離心管中,加入8 m L Middlebrook7H9液體培養(yǎng)基,再接種初始濃度為1010CFU/m L的MTB懸液2 m L混勻,恒溫(37℃)下培養(yǎng)30 d,從每支離心管中,隨機(jī)取出一塊材料,經(jīng)固定、干燥、噴金后,應(yīng)用掃描電子顯微鏡觀察,比較MTB對(duì)不同植入材料的黏附力,同時(shí)觀察界面生物膜形成情況。然后每支離心管中加入2 m L濃度為1μg/m L的利福平溶液,同樣條件下繼續(xù)培養(yǎng)30 d。取出材料塊,經(jīng)固定、干燥、噴金后,應(yīng)用掃描電子顯微鏡觀察,了解利福平對(duì)MTB黏附力的影響及對(duì)MTB生物膜的破壞情況。結(jié)果 MTB對(duì)不同植入材料界面的黏附力由強(qiáng)到弱依次是聚乙烯、鈷鉻鉬、鈦合金;聚乙烯界面可見(jiàn)生物膜形成,鈷鉻鉬合金、鈦合金界面均未見(jiàn)生物膜形成;利福平干預(yù)后,植入物界面黏附的菌落數(shù)顯著減少,聚乙烯界面生物膜呈不同程度破壞。結(jié)論利福平能抑制MTB對(duì)不同植入材料界面的黏附及生物膜形成。
