【摘要】：目的觀察血吸蟲活蟲卵在不同保存液中的保存時(shí)間及孵化毛蚴的活力,獲得血吸蟲活蟲卵最佳保存條件,探索用作孵化參考品的可行性。方法收集人工感染血吸蟲家兔直腸糞便,經(jīng)80、100、160孔/25.4 mm和200孔/25.4 mm分樣篩過濾去除粗糞渣,260孔/25.4 mm收集血吸蟲活蟲卵,于0.9%Na Cl溶液、1.2%Na Cl溶液、磷酸鹽緩沖液(PBS,p H7.2)、1.0%蔗糖溶液和Mili-Q水5種保存液中混勻,分別于4℃和15℃恒溫保存。每隔7 d對保存蟲卵進(jìn)行孵化,觀察毛蚴活力并計(jì)數(shù)。結(jié)果在4℃條件下,1.0%蔗糖溶液和1.2%Na Cl溶液中蟲卵孵化率分別于49 d后和126 d后降至0;0.9%Na Cl溶液和PBS保存液中蟲卵孵化率于112 d后降至10%以下,140 d后孵出的毛蚴活力較差;Mili-Q水保存的蟲卵孵化率于196 d后降至10%以下,280 d后孵出毛蚴,活力明顯下降且不利于觀察。在15℃條件下,各保存液中蟲卵的孵化率分別在49~105 d之間逐漸降至0。結(jié)論實(shí)驗(yàn)室以Mili-Q水為血吸蟲活蟲卵保存液,置4℃恒溫保存,196 d之內(nèi)的冷藏蟲卵具有用作孵化陽性參考品的價(jià)值。
[Abstract]:Objective to observe the preservation time of live eggs of Schistosoma japonicum and the activity of hatching cercariae in different preservation solution, to obtain the best preservation conditions of live eggs of Schistosoma japonicum, and to explore the feasibility of using them as reference materials for hatching.
