分枝桿菌噬菌體在抗結(jié)核研究中的作用

【摘要】:分子桿菌噬菌體是其宿主分枝桿菌的病毒,前者對于結(jié)核病控制及臨床結(jié)核菌診斷有良好的應(yīng)用前景。目前已有超過1000株分枝桿菌噬菌體進(jìn)行了全基因組測序,結(jié)果顯示分枝桿菌噬菌體基因組具有豐富的多樣性和鑲嵌結(jié)構(gòu),且都編碼有噬菌體裂解酶。裂解酶是噬菌體在感染宿主末期表達(dá)的蛋白,其能水解細(xì)胞壁以釋放子代噬菌體。除裂解酶外,分枝桿菌噬菌體還編碼其它有抗結(jié)核活性的多肽及蛋白。分枝桿菌噬菌體在臨床耐藥結(jié)核菌的檢測中也發(fā)揮了巨大作用。本文綜述了來源于分枝桿菌噬菌體的多肽與蛋白的抗結(jié)核活性,并系統(tǒng)闡述了分枝桿菌噬菌體在臨床結(jié)核菌耐藥性檢測中的應(yīng)用,并展望了工程噬菌體在殺死耐藥病原菌研究中的應(yīng)用前景。
[Abstract]:Molecular bacteriophage is the host virus of Mycobacterium spp. The former has a good application prospect for tuberculosis control and clinical diagnosis of tuberculous bacilli. Up to now, more than 1000 strains of Mycobacterium phage have been sequenced. The results show that the phage genome of Mycobacterium has rich diversity and mosaic structure, and all of them encode phage lyase. Lyase is a protein expressed by phage at the end of infected host. It can hydrolyze cell wall to release offspring phage. Besides lyase, Mycobacterium phage also encodes other peptides and proteins with anti-tuberculosis activity. Mycobacterium bacteriophage also plays an important role in the detection of drug-resistant tuberculous bacteria. In this paper, the antituberculous activity of peptides and proteins from Mycobacterium phage was reviewed, and the application of Mycobacterium phage in the detection of drug resistance of Mycobacterium tuberculosis was systematically reviewed. The application prospect of engineering bacteriophage in killing resistant pathogenic bacteria was prospected.
