血漿miR-122在抗-HCV陽性獻(xiàn)血者中表達(dá)的動態(tài)分析

【摘要】:目的動態(tài)分析抗-HCV陽性獻(xiàn)血者血漿miR-122的表達(dá)特點及與HCV感染診斷指標(biāo)的關(guān)聯(lián)。方法對133名抗-HCV陽性獻(xiàn)血者、24名健康獻(xiàn)血者及15名確證抗-HCV陽性追蹤獻(xiàn)血者血液標(biāo)本抽提血漿miRNA,以實時熒光定量PCR法檢測血漿miR-122,分析血漿miR-122相對表達(dá)量與HCV RNA、抗-HCV臨界值指數(shù)(COI)等指標(biāo)關(guān)聯(lián)性。結(jié)果 ROC曲線分析:miR-122在抗-HCV+/HCV RNA+組和抗-HCV+/HCV RNA-組中診斷丙型肝炎的AUC(ROC曲線下面積)為0.830(95%CI:0.755-0.889),區(qū)分抗-HCV+/HCV RNA+組和抗-HCV+/HCV RNA-組的敏感性和特異性分別為74.7%和82.4%。追蹤15例抗-HCV+/HCV RNA+獻(xiàn)血者血漿miR-122表達(dá)明顯降低(P0.05)。結(jié)論血漿miR-122與HCV感染密切相關(guān),確證HCV RNA感染獻(xiàn)血者血漿miR-122隨時間遷移表達(dá)降低。
[Abstract]:Objective to dynamically analyze the expression of plasma miR-122 in anti-HCV positive blood donors and its association with HCV infection diagnosis. Methods Plasma miRNA, was extracted from 133 blood samples of anti HCV positive blood donors, 24 healthy blood donors and 15 confirmed anti HCV positive blood donors. The real-time fluorescence quantitative PCR method was used to detect plasma miR-122,. The correlation between the relative expression of plasma miR-122 and the critical value index (COI) of HCV RNA, anti-HCV was analyzed. Results ROC curve analysis: the area under AUC (ROC curve of miR-122 in anti HCV / HCV RNA group and anti HCV / HCV RNA- group was 0.830 (95%CI:0.755-0.889). The sensitivity and specificity of differentiating anti-HCV / HCV RNA and anti-HCV / HCV RNA- were 74.7% and 82.4%, respectively. The expression of plasma miR-122 was significantly decreased in 15 anti-HCV / HCV RNA donors (P 0.05). Conclusion there is a close relationship between plasma miR-122 and HCV infection. It is confirmed that the expression of plasma miR-122 decreases with time migration in HCV RNA infected blood donors.
