【摘要】：目的表達和純化布魯氏菌OMP25重組蛋白并制備OMP25多克隆抗體,為建立布魯氏菌病的診斷方法奠定基礎(chǔ)。方法采用PCR方法擴增M16羊布魯氏菌標(biāo)準(zhǔn)菌株的OMP25外膜蛋白基因,連接載體質(zhì)粒pET-30a(+)并導(dǎo)入受體細胞DH5α中,構(gòu)建重組PET-30a(+)-OMP25質(zhì)粒,轉(zhuǎn)化至Rosetta(DE3)菌株,收集表達的蛋白,用Ni2+NTA柱純化試劑盒進行純化。用純化的重組蛋白免疫家兔,制備OMP25多克隆抗體,Western blot檢測抗體特異性。結(jié)果成功構(gòu)建重組質(zhì)粒PET-30a(+)-OMP25,導(dǎo)入受體細胞Rosetta(DE3),獲得重組蛋白,純化后免疫家兔,制備的OMP25多克隆抗體與OMP25重組蛋白發(fā)生特異性反應(yīng)。結(jié)論成功構(gòu)建重組質(zhì)粒PET-30a(+)-OMP25,表達的重組蛋白具有免疫原性,并制備了高純度、高效價的多克隆抗體,為布病免疫診斷方法的建立奠定了基礎(chǔ)。
[Abstract]:Objective to express and purify the recombinant protein of brucellosis OMP25 and to prepare OMP25 polyclonal antibody, so as to establish a diagnostic method for brucellosis. Methods the OMP25 outer membrane protein gene of standard strain of Brucella sheep M16 was amplified by PCR. The plasmid pET-30a () was ligated into the receptor cell DH5 偽. The recombinant PET-30a () -OMP25 plasmid was constructed and transformed into Rosetta (DE3) strain. The expressed protein was purified by Ni2 NTA column purification kit. OMP25 polyclonal antibody (, Western blot) was prepared by immunizing rabbits with purified recombinant protein. Results Recombinant plasmid PET-30a ()-OMP25, was successfully constructed and transfected into receptor cell Rosetta (DE3). The recombinant protein was purified and immunized rabbits. The OMP25 polyclonal antibody was specifically reacted with OMP25 recombinant protein. Conclusion the recombinant protein expressed by recombinant plasmid PET-30a ()-OMP25, has immunogenicity, and the polyclonal antibody with high purity and high titer has been prepared, which lays a foundation for the establishment of immune diagnosis method for brucellosis.
