【摘要】：目的:探討Dectin-1在熱處理光滑念珠菌刺激大鼠氣道上皮細(xì)胞(RTECs)中的作用及對(duì)IL-10和TNF-α表達(dá)的影響。方法:將體外培養(yǎng)RTECs隨機(jī)分為三組:對(duì)照組加無(wú)菌生理鹽水;真菌刺激組加熱處理光滑念珠菌孵育;抑制劑干預(yù)組在昆布多糖預(yù)處理后加熱處理光滑念珠菌孵育。分別在孵育0h、2h、4h、6h后終止,收集細(xì)胞及上清液。在光鏡下觀察細(xì)胞形態(tài)及生長(zhǎng)情況,用MTT法檢測(cè)細(xì)胞存活率,Western blot法檢測(cè)Dectin-1蛋白的表達(dá),ELISA法檢測(cè)IL-10和TNF-α的表達(dá)。結(jié)果:熱處理光滑念珠菌抑制RTECs的增殖;真菌刺激組、抑制劑干預(yù)組2h、4h、6h細(xì)胞Dectin-1、IL-10、TNF-α的表達(dá)均高于對(duì)照組(P均0.05)。在抑制劑干預(yù)組,同時(shí)間點(diǎn)Dectin-1、IL-10、TNF-α的表達(dá)均低于真菌刺激組(P0.05)。結(jié)論:Dectin-1是RTECs識(shí)別熱處理光滑念珠菌的重要受體,并誘導(dǎo)其釋放IL-10和TNF-α,介導(dǎo)炎癥反應(yīng)的發(fā)生。Dectin-1抑制劑昆布多糖能抑制IL-10和TNF-α的表達(dá),加重機(jī)體損傷。
[Abstract]:Aim: to investigate the effect of Dectin-1 on the expression of IL-10 and TNF- 偽 in cultured rat airway epithelial cells stimulated by Candida smoothing. Methods: RTECs cultured in vitro was randomly divided into three groups: control group with aseptic saline; fungal stimulation group with heating treated Candida smooth; inhibitor intervention group with heat treated Candida smoothing after pretreatment with laminin polysaccharide. The cells and supernatant were collected after incubation for 2 h and 4 h respectively for 6 h. The morphology and growth of cells were observed under light microscope. The expression of Dectin-1 protein was detected by MTT method, and the expression of IL-10 and TNF- 偽 was detected by ELISA method. Results: heat treated Candida smoothing inhibited the proliferation of RTECs, and the expression of Dectin-1,IL-10,TNF- 偽 in fungal stimulation group and inhibitor intervention group was significantly higher than that in control group (P 0.05). In the inhibitor intervention group, the expression of Dectin-1,IL-10,TNF- 偽 at the same time was lower than that in the fungal stimulation group (P0.05). Conclusion: Dectin-1 is an important receptor for RTECs to recognize heat treated Candida albicans, and to induce the release of IL-10 and TNF- 偽, thus mediating inflammation. Dectin-1 inhibitor laminin polysaccharide can inhibit the expression of IL-10 and TNF- 偽 and aggravate the damage of organism.
