【摘要】：目的：探討Toll樣受體4基因在HIV相關(guān)型KS (Kaposi's sarcoma)腫瘤組織和及正常組織中的表達(dá)。方法：收集2011年5月至2013年7月在新疆醫(yī)科大學(xué)第一附屬醫(yī)院及新疆傳染病醫(yī)院就診并經(jīng)病理明確診斷的患者KS腫瘤組織35例,包括HIV-KS26例,經(jīng)典KS9例,應(yīng)用實(shí)時(shí)熒光定量PCR技術(shù)、免疫組化方法檢測(cè)TLR4在卡波西肉瘤組織中的表達(dá)與分布。應(yīng)用Western Blot方法檢測(cè)TLR4信號(hào)通路上蛋白的表達(dá)。結(jié)果：TLR4基因在KS腫瘤組織中mRNA水平的表達(dá)均低于正常組織,差異有統(tǒng)計(jì)學(xué)意義(P=0.0017),經(jīng)典KS及正常對(duì)照之間表達(dá)有顯著性差異(P=0.0497), HIV-KS與正常對(duì)照之間表達(dá)也有顯著性差異(P=0.0054)。而在經(jīng)典KS及HIV-KS之間表達(dá)無統(tǒng)計(jì)學(xué)意義(P=0.46)。免疫組化中,在經(jīng)典KS及HIV-KS病損皮膚組織真皮層可見TLR4陽性細(xì)胞,主要表達(dá)于內(nèi)皮細(xì)胞和異形細(xì)胞胞漿和胞膜。蛋白水平：HIV-KS及經(jīng)典KS組織中可見到Ras、Raf、ERK、IκB-α、P50、P65表達(dá)較正常組織高,P0.05,且有顯著性差異,而在]HIV-KS和經(jīng)典KS之間無顯著性差異。其余蛋白在正常及KS之間表達(dá)無顯著性差異。結(jié)論：在艾滋病患者中,TLR4低表達(dá)與HIV-KS的發(fā)生密切相關(guān)。
[Abstract]:Aim: to investigate the expression of Toll-like receptor 4 gene in HIV-associated KS (Kaposi's sarcoma) tumors and normal tissues. Methods: from May 2011 to July 2013, 35 patients with KS tumor tissue, including HIV-KS26, classic KS9, were collected from the first affiliated Hospital of Xinjiang Medical University and Xinjiang Infectious Diseases Hospital. The expression and distribution of TLR4 in Kaposi's sarcoma were detected by real-time fluorescence quantitative PCR and immunohistochemistry. Western Blot method was used to detect the expression of protein in TLR4 signaling pathway. Results: the expression of TLR4 gene in KS was significantly lower than that in normal tissue (P0. 0017). There was a significant difference between classical KS and normal control (P0. 0497). There was also a significant difference between HIV-KS and normal control (P0. 0054). However, there was no significant difference between the expression of KS and HIV-KS (P < 0. 46). In immunohistochemistry, TLR4 positive cells were found in the dermis of classical KS and HIV-KS lesions, mainly expressed in the cytoplasm and membrane of endothelial cells and allogeneic cells. Protein level: the expression of Ras,Raf,ERK,I 魏 B- 偽 and P50 p65 in HIV-KS and classical KS tissues was higher than that in normal tissues, and there was significant difference between HIV-KS and classical KS, but there was no significant difference between HIV-KS and classical KS. There was no significant difference in the expression of other proteins between normal and KS. Conclusion: the low expression of TLR4 is closely related to the occurrence of HIV-KS in AIDS patients.
