【摘要】：目的探討檢測抗日本血吸蟲三磷酸甘油酸脫氫酶(Sj GAPDH)抗體IgG診斷血吸蟲病的價(jià)值。方法收集血吸蟲現(xiàn)癥感染患者治療前、后不同時間點(diǎn)的血清及其他寄生蟲病患者血清。以純化的重組Sj GAPDH蛋白為抗原建立檢測抗Sj GAPDH抗體IgG的常規(guī)酶聯(lián)免疫吸附試驗(yàn)方法(Sj GAPDH ELISA)及生物素-親和素信號放大酶聯(lián)免疫吸附試驗(yàn)方法(Sj GAPDH BA ELISA),同時對收集的血清標(biāo)本進(jìn)行抗Sj GAPDH抗體IgG檢測,并與檢測抗SEA抗體IgG的SEA ELISA結(jié)果作比較,評價(jià)檢測抗Sj GAPDH抗體IgG用于血吸蟲病診斷與療效考核的價(jià)值。結(jié)果 Sj GAPDH ELISA和Sj GAPDH BA ELISA檢測79份現(xiàn)癥感染血吸蟲患者血清抗Sj GAPDH抗體IgG的敏感性分別為54.43%和91.14%,差異有統(tǒng)計(jì)學(xué)意義(P0.05%);特異性分別為93.85%和92.31%,差異無統(tǒng)計(jì)學(xué)意義(P0.05%)。Sj GAPDH ELISA檢測患者在治療后6個月血清特異性抗體陰轉(zhuǎn)率為44.19%,治療后12個月陰轉(zhuǎn)率達(dá)53.49%。Sj GAPDH BA ELISA檢測患者治療后6個月血清特異性抗體陰轉(zhuǎn)率為5.55%,治療后12個月陰轉(zhuǎn)率為9.72%�；颊哐逯锌筍EA抗體IgG治療后6和12個月均陽性。結(jié)論 Sj GAPDH蛋白在患者體內(nèi)誘導(dǎo)短壽抗體反應(yīng),檢測患者血清中的抗Sj GAPDH特異性抗體IgG水平及其在治療前、后的變化具有一定的診斷與療效考核價(jià)值。
[Abstract]:Objective to investigate the value of detecting (Sj GAPDH) antibody against triphosphate dehydrogenase (Sj GAPDH) in the diagnosis of schistosomiasis japonicum. Methods Serum samples of patients with schistosomiasis were collected at different time points before and after treatment. Using purified Recombinant Sj GAPDH protein as Antigen to establish routine Enzyme-Linked Immunosorbent Assay for Detection of Antibody to Sj GAPDH IgG (Sj GAPDH ELISA) and (Sj GAPDH BA ELISA), for Biotin / Affinity signal Amplification Enzyme-linked Immunosorbent Assay To detect the anti Sj GAPDH antibody IgG. Compared with the SEA ELISA results of detecting anti SEA antibody IgG, the value of detecting anti Sj GAPDH antibody IgG for schistosomiasis diagnosis and curative effect evaluation was evaluated. Results the sensitivity of Sj GAPDH ELISA and Sj GAPDH BA ELISA in detecting anti-Sj GAPDH antibody IgG were 54.43% and 91.14%, respectively (P0.05%), the specificity was 93.85% and 92.31%, respectively (P0.05%). Sj GAPDH). The seroconversion rate of serum specific antibody was 44.19 in 6 months after ELISA treatment, 5.55 in 6 months after treatment and 9.72 in 12 months after treatment. The negative conversion rate of serum specific antibody was 5.55% at 12 months after treatment, 5.55% at 6 months after treatment and 9.72% at 12 months after treatment. Serum anti SEA antibody IgG was positive 6 and 12 months after treatment. Conclusion Sj GAPDH protein induces short life antibody reaction in patients, and the detection of IgG level of anti Sj GAPDH specific antibody in patients' serum and its changes before and after treatment have certain diagnostic and therapeutic value.
