【摘要】：目的探討乙型肝炎(乙肝)病毒感染對(duì)ABCB4基因的影響以及與膽囊膽固醇結(jié)石形成的相關(guān)性。方法選擇行膽囊切除的膽囊結(jié)石患者48例,HBs Ag陽性28例為觀察組,HBs Ag陰性20例為對(duì)照組。兩組膽囊分別做HE染色及免疫組化S-P法檢測(cè)乙肝病毒表面抗原和核心抗原,應(yīng)用Real-time PCR技術(shù)檢測(cè)兩組的ABCB4基因表達(dá)水平。結(jié)果 HBs Ag在膽囊黏膜組織中的表達(dá),觀察組陽性率為57.1%,對(duì)照組全為陰性,兩組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。HBc Ag在膽囊黏膜中的表達(dá),觀察組陽性率為28.6%,對(duì)照組全為陰性,兩組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。觀察組ABCB4基因mRNA表達(dá)(0.909±0.059)較低,對(duì)照組ABCB4基因mRNA表達(dá)(1.149±0.039)較高,兩組比較差異有統(tǒng)計(jì)學(xué)意義(t=-11.062,P0.05)。結(jié)論乙肝病毒感染患者膽囊ABCB4基因的表達(dá)下調(diào),可能導(dǎo)致膽囊膽固醇結(jié)石的形成。
[Abstract]:Objective to investigate the effect of hepatitis B virus infection on ABCB4 gene and its correlation with cholesterol gallstone formation. Methods A total of 48 patients with cholecystolithiasis underwent cholecystectomy were divided into two groups: control group (n = 20). Hepatitis B virus surface antigen and core antigen were detected by HE staining and S-P immunohistochemical method. ABCB4 gene expression was detected by Real-time PCR technique. Results the positive rate of HBs Ag in gallbladder mucosa was 57.1 in the observation group and negative in the control group. The difference between the two groups was statistically significant (P0.05). The positive rate of HBc Ag in the observation group was 28.6g and that in the control group was negative. The difference between the two groups was statistically significant (P0.05). The mRNA expression of ABCB4 gene in the observation group (0.909 鹵0.059) was lower than that in the control group (1.149 鹵0.039). The difference between the two groups was statistically significant (P0.05). Conclusion the down-regulated expression of ABCB4 gene may lead to the formation of cholecystolithiasis in patients with hepatitis B virus infection.
