不同滴度噬菌體D29對氣道上皮細(xì)胞9HTE的生長及功能影響研究

【摘要】:目的研究噬菌體D29不同滴度對氣道上皮細(xì)胞株9HTE的生長及功能影響。方法分別用高滴度(109PFU/mL)和低滴度(107 PFU/mL)噬菌體D29及噬菌體緩沖液組作用于9HTE細(xì)胞株后,用四甲基偶氮唑藍(lán)比色法(MTT)檢測細(xì)胞生長情況;用酶聯(lián)免疫吸附試驗(ELISA)法測量細(xì)胞上清液白細(xì)胞介素(IL)-6、IL-8水平;用逆轉(zhuǎn)錄PCR(RT-PCR)檢測細(xì)胞間黏附分子-1(ICAM-1)mRNA的表達(dá),用流式細(xì)胞技術(shù)檢測細(xì)胞凋亡率。結(jié)果高、低滴度噬菌體D29作用氣道上皮細(xì)胞一段時間后,對細(xì)胞生長活性、細(xì)胞因子IL-6、IL-8和ICAM-1mRNA的表達(dá),以及細(xì)胞凋亡率的影響與噬菌體緩沖液組比較差異無統(tǒng)計學(xué)意義(P0.05)。結(jié)論高、低滴度噬菌體D29對體外培養(yǎng)的氣道上皮細(xì)胞生長及功能無明顯影響。
[Abstract]:Objective to study the effects of different titers of phage D 29 on the growth and function of airway epithelial cell line 9 HTE. Methods High titer (109 PFU / mL) and low titer (107 PFU / mL) phage D29 and bacteriophage buffer group were used to treat 9HTE cell line respectively. The cell growth was detected by MTT assay. The levels of interleukin-6 (IL-8) in supernatant were measured by enzyme-linked immunosorbent assay (Elisa), the expression of intercellular adhesion molecule-1 (ICAM-1) mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR), and the apoptosis rate was detected by flow cytometry. Results the effects of high and low titer phage D29 on the cell growth activity, the expression of cytokines IL-6, IL-8 and ICAM-1 mRNA, and the rate of apoptosis were not significantly different from those of phage buffer group (P0.05). Conclusion High and low titer phage D 29 has no effect on the growth and function of cultured airway epithelial cells.
