抑制Kupffer細胞功能減輕日本血吸蟲病肝臟肉芽腫炎癥反應(yīng)

【摘要】:目的通過制備氯磷酸二鈉脂質(zhì)體(LE-Cl2MDP),選擇性剔除日本血吸蟲感染小鼠模型中Kupffer細胞,研究抑制Kupffer細胞功能對日本血吸蟲肝臟肉芽腫形成的影響。方法采用脂質(zhì)體包裹CL2MDP;6~8周齡雌性BALB/c小鼠分為3組:PBS組,日本血吸蟲感染組,日本血吸蟲感染聯(lián)合LE-Cl2BMP組;對于聯(lián)合組,小鼠尾靜脈注射LE-Cl2MDP(0.1mL/10g),每周2次,連續(xù)6周;感染后第12周,剝殺小鼠,留取血清及肝臟,用于細胞因子和免疫組織化學(xué)檢測等,數(shù)據(jù)統(tǒng)計分析。結(jié)果經(jīng)過LE-Cl2MDP處理后,日本血吸蟲感染肉芽腫周圍Kupffer細胞細胞明顯減少;無論是感染第8周,還是第12周肝組織中蟲卵肉芽腫周圍炎癥細胞浸潤明顯減少,纖維化程度減輕;同時降低感染第12周小鼠血清IL-10水平,LE-Cl2MDP組為(42.6±10.4)pg/mL,感染組為(67.4±12.9)pg/mL(P0.05)。結(jié)論本研究建立LE-Cl2MDP剔除小鼠肝臟Kupffer細胞的日本血吸蟲感染模型,抑制了肝組織中蟲卵肉芽腫周圍炎癥反應(yīng)。
[Abstract]:Objective to study the effect of inhibition of Kupffer cell function on the formation of hepatic granuloma of Schistosoma japonicum by preparing LE-Cl2MDP and selectively culling Kupffer cells from Schistosoma japonicum (Schistosoma japonicum) model. Methods female BALB / c mice were divided into three groups by liposome-encapsulated CL2MDP 6week-old female BALB / c mice into three groups: Schistosoma japonicum infection group and LE-Cl2BMP group; for the combined group, mice were injected with LE-Cl2MDP (0.1 mL / 10 g) twice a week, for 6 weeks; 12 weeks after infection, the mice were treated with LE-Cl2MDP (0.1 mL / 10 g) twice a week. The mice were stripped, serum and liver were retained for cytokine and immunohistochemical detection, and statistical analysis of data was made. Results after treatment with LE-Cl2MDP, the Kupffer cells around the granuloma infected by Schistosoma japonicum decreased significantly, and the infiltration of inflammatory cells around the egg granuloma decreased significantly and the degree of fibrosis decreased in the liver tissue at the 8th and 12th week of infection. At the same time, the serum IL-10 level was decreased in LE-Cl2MDP group (42.6 鹵10.4) PG / mL, and in the infected group was (67.4 鹵12.9) PG / mL (P0.05). Conclusion the model of Schistosoma japonicum infection with Kupffer cells in liver of LE-Cl2MDP knockout mice was established and the inflammatory reaction around egg granuloma in liver tissue was inhibited.
