徐州地區(qū)結(jié)核分枝桿菌基因分型及其與利福平耐藥基因突變的關(guān)系

【摘要】:目的探究徐州地區(qū)結(jié)核分枝桿菌的基因型分布特征及其與利福平耐藥基因突變形成的關(guān)系。方法選取徐州市傳染病醫(yī)院結(jié)核病實驗室基因型毒株500株,其中含有經(jīng)羅氏比例法鑒定的耐利福平臨床分離株300株和利福平敏感200株,采用間隔區(qū)寡核苷酸分型(spoligotyping)分析其基因型結(jié)構(gòu)及其與利福平抗性基因的突變關(guān)系。結(jié)果徐州地區(qū)的結(jié)核分枝桿菌的基因分型主要為北京家族基因群(61.2%)和非北京家族基因群(38.8%),利福平耐藥和敏感組北京家族基因群的構(gòu)成比差異無統(tǒng)計學(xué)意義(醊2=0.09,P0.05);北京家族531位點、516位點以及531和526位點突變率分別為55.49%、10.99%和7.69%與非北京家族33.90%、21.19%和15.25%相比,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論徐州地區(qū)結(jié)核分枝桿菌基因型為北京家族基因群和非北京家族基因群,非北京家族基因群具有基因多態(tài)性,兩者與利福平耐藥率無明顯相關(guān),兩者在單531、單516以及531和526利福平耐藥位點突變率存在差異。
[Abstract]:Objective to investigate the genotypic distribution of Mycobacterium tuberculosis in Xuzhou and its relationship with rifampicin resistance gene mutation. Methods 500 strains of genotypic strains of tuberculosis were selected from Xuzhou Infectious Disease Hospital, including 300 strains of rifampin resistant clinical isolates and 200 strains of rifampicin sensitive strains identified by Roche scale method. The genotype structure and its relationship with rifampicin resistance gene were analyzed by spacer oligonucleotide typing (spoligotyping). Results the main genotypes of Mycobacterium tuberculosis in Xuzhou area were Beijing family gene group (61.2%) and non-Beijing family gene group (38.8%). There was no significant difference between rifampicin resistance group and sensitive group (P 0.05). The mutation rates of 531, 531 and 526 loci in Beijing family were 55.49% and 7.69%, respectively, compared with 33.90% and 15.25% in non-Beijing families. The difference was statistically significant (P0.05). Conclusion the genotypes of Mycobacterium tuberculosis in Xuzhou area are Beijing family gene group and non-Beijing family gene group. The gene polymorphism of non-Beijing family gene group has no significant correlation with rifampin resistance rate. The mutation rates of rifampin resistance loci in single 531, single 516 and 531 and 526 rifampicin were different between the two groups.
