貴州省遵義地區(qū)人體結(jié)膜吸吮線蟲線粒體DNA分析

【摘要】:目的通過線粒體DNA測序及親緣關(guān)系分析初步確定貴州省遵義地區(qū)結(jié)膜吸吮線蟲的來源。方法采集2016年貴州省遵義地區(qū)7例人體結(jié)膜吸吮線蟲病例患者中取出的蟲體,提取DNA,PCR擴(kuò)增結(jié)膜吸吮線蟲線粒體DNA COX1序列,與已發(fā)表的結(jié)膜吸吮線蟲線粒體DNA COX1序列進(jìn)行比對和系統(tǒng)進(jìn)化分析。結(jié)果自結(jié)膜吸吮線蟲患者所得蟲體擴(kuò)增的COX1基因存在差異,人體感染的蟲株之間存在較低的遺傳變異。結(jié)論遵義地區(qū)是新的結(jié)膜吸吮線蟲病流行區(qū),其結(jié)膜吸吮線蟲流行株包含了亞洲及歐洲兩種來源,至少有4種單倍體型。
[Abstract]:Objective to determine the origin of conjunctival sucking nematode in Zunyi area of Guizhou province by mitochondrial DNA sequencing and phylogenetic analysis. Methods A total of 7 cases of human conjunctival sucking nematode were collected from Zunyi district of Guizhou province in 2016, and DNA PCR was extracted to amplify the COX1 sequence of mitochondrial DNA in conjunctival sucking nematode. Alignment and phylogenetic analysis of COX1 sequence of mitochondrial DNA of nematode conjunctiva were carried out. Results there were differences in the COX1 gene amplified from the worm from the patients with sucking nematode conjunctiva, and there was a low genetic variation among the infected strains. Conclusion Zunyi area is a new endemic area of conjunctival sucking nematode.
