綠原酸、異綠原酸對煙曲霉菌生物被膜抑制作用的體外研究

【摘要】:目的:通過在體外復(fù)制煙曲霉菌(Aspergillus fumigates)的生物被膜(Biofilm, BF)模型,研究綠原酸(Chlorogenic acid,CRA)、異綠原酸(Isochlorogenic acid,IRC)對生物被膜的影響。 方法:選取臨床分離的煙曲霉菌構(gòu)建體外生物被膜模型,微量液基稀釋法測定最低抑菌濃度(Minimal inhibition concentration, MIC),,將菌株分為空白對照組、實驗組(CRA1024μg/ml、CRA512μg/ml、CRA256μg/ml、IRC1000μg/ml、IRC500μg/ml、IRC250μg/ml),掃描電子顯微鏡(Scanningelectron microscopy,SEM)、激光共聚焦掃描顯微鏡(Laser confocal scanningmicroscope, CLSM)定性觀察BF;結(jié)晶紫染色法半定量生物被膜量。 結(jié)果:37℃培養(yǎng)24h煙曲霉菌形成早期BF,48h形成成熟期BF。實驗組煙曲霉菌BF均較其相應(yīng)空白對照組空洞、稀疏,但菌絲未見明顯減少,死活菌數(shù)也無明顯差別。CRA組早期結(jié)晶紫半定量結(jié)果分別為(1.05±0.19)、(1.14±0.26)、(1.16±0.14),IRC組為(1.18±0.06)、(1.21±0.06)、(1.26±0.04),均少于空白對照組(1.91±0.17),差異具有統(tǒng)計學意義(P0.05);CRA組成熟期結(jié)晶紫半定量結(jié)果為(2.25±0.05)、(2.27±0.05)、(2.31±0.03),IRC組為(2.26±0.02)、(2.27±0.02)、(2.29±0.04),也均少于空白對照組(2.36±0.01),差異具有統(tǒng)計學意義(P0.05)。 結(jié)論:1、可以在體外建立煙曲霉菌生物被膜模型;2、CRA、IRC對體外煙曲霉菌BF形成有抑制作用。
[Abstract]:Objective: to study the effect of Lv Yuan acid Chlorogenic acidoglycan (Isochlorogenic acidogenic) on biofilm by replicating the biofilm model of Aspergillus fumigatesin vitro. Methods: a biofilm model was established in vitro by clinical isolates of Aspergillus fumigates.Methods: the biofilm model of Aspergillus fumigatesin vitro was established. Minimal inhibition concentration was determined by microdilution method. The strains were divided into blank control group. CRA1024 渭 g / ml CRA512 渭 g / ml CRA256 渭 g / ml IRC1000 渭 g / ml IRC500 渭 g / ml IRC250 渭 g / ml, scanning electron microscope (SEM), laser confocal scanning microscope (laser confocal scanning microscope), laser confocal scanning microscope (laser confocal scanning microscope), laser scanning microscope (CLSM), and semi-quantitative biofilm quantity of CRA 256 渭 g / ml.
