【摘要】:目的 :分析新發(fā)展晚期血吸蟲病患者血清多肽,篩選有潛在應(yīng)用價值的標(biāo)志蛋白,為其診斷與發(fā)病機制研究奠定基礎(chǔ)。方法:采集新發(fā)展晚期血吸蟲病患者與健康人血清,利用包被有弱陽離子的磁珠富集純化血清多肽,應(yīng)用基質(zhì)輔助激光解析電離時間飛行質(zhì)譜(MALDI-TOF-MS)Flex分析技術(shù)結(jié)合Mascot生物信息學(xué)分析血清多肽指紋圖譜。結(jié)果:與健康對照組比較,在0.8~13.0 ku區(qū)段,新發(fā)展晚期血吸蟲病患者血清中m/z 2 082、4 282多肽表達(dá)上調(diào)(P0.01),2 661、2 991、3 241、5 337、5 906多肽表達(dá)下調(diào)(P0.05);常規(guī)晚期血吸蟲病患者血清中m/z 2 662、4 209、5 337與5 906處顯著下調(diào),2 082處顯著上調(diào)。結(jié)論:m/z3 241、4 282血清差異多肽對新發(fā)展晚期血吸蟲病患者的診斷可能具有潛在應(yīng)用價值,對該病發(fā)生發(fā)展的機制也奠定了理論基礎(chǔ)。
[Abstract]:Objective: to analyze the serum polypeptides of patients with advanced schistosomiasis and to screen potential marker proteins for the diagnosis and pathogenesis of schistosomiasis. Methods: serum samples of newly developed advanced schistosomiasis patients and healthy persons were collected and purified by magnetic beads coated with weak cations. Matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS Flex) was used to analyze serum polypeptide fingerprints with Mascot bioinformatics. Results: compared with the healthy control group, in the region of 0.8 ~ 13.0 ku, In the serum of newly developed advanced schistosomiasis patients, the expression of m / z 2 082n 4 282 polypeptide was up-regulated, and the expression of m- / z 2662 + 4209H 5 337 and 5 906 was significantly down-regulated at 2 082 and 2 082 points respectively in the serum of newly developed patients with advanced schistosomiasis, and the expression of P0.05906 polypeptide was down-regulated at 3241U 5337N 5906, and that in the serum of patients with advanced schistosomiasis was significantly lower than that in the patients with advanced schistosomiasis. ConclusionThe differential polypeptide in serum of w / m / z _ 3 241N _ 4 282 may have potential application value in the diagnosis of newly developed advanced schistosomiasis, and it also lays a theoretical foundation for the pathogenesis of schistosomiasis.
