PI4KA與乙型肝炎病毒復(fù)制的相關(guān)性研究

【摘要】:目的:通過(guò)探討PI4KA與慢性乙型肝炎患者病毒載量的關(guān)系;觀察PI4KA在肝癌細(xì)胞株HepG2及HepG2.2.15中表達(dá)的差異,探究HBV對(duì)PI4KA表達(dá)的影響;在臨床和細(xì)胞水平探討PI4KA與乙型肝炎病毒復(fù)制的關(guān)系。方法:收集180例慢性乙型肝炎患者的血清,利用熒光定量PCR檢測(cè)其HBV DNA的載量,根據(jù)其HBV DNA載量的高低,分為三組:高病毒載量組(HBV DNA1×107拷貝/m1)、中病毒載量組(1×105拷貝/ml≤HBV DNA≤1×107拷貝/m1)及低病毒載量組(]3BV DNA1×105拷貝/m1),同時(shí)以60名健康人作對(duì)照。采用ELISA法,檢測(cè)各組外周血清中的PI4KA的含量;培養(yǎng)HepG2及HepG2.2.15細(xì)胞,當(dāng)細(xì)胞融合達(dá)到90%時(shí),提取兩細(xì)胞RNA并逆轉(zhuǎn)錄成cDNA,采用實(shí)時(shí)熒光定量PCR法檢測(cè)HepG2和HepG2.2.15細(xì)胞中PI4KA mRNA的表達(dá)水平;采用ELISA法檢測(cè)此兩種細(xì)胞培養(yǎng)上清液中PI4KA的表達(dá)水平。 結(jié)果:60例健康人血清PI4KA的含量為2.29±0.75ng/ml;60例低病毒載量組血清PI4KA的含量為2.73±0.71ng/ml;60例中病毒載量組血清PI4KA的含量為3.52±0.78ng/ml及60例高病毒載量組血清PI4KA含量為4.72±0.77ng/ml。慢性乙型肝炎患者不同病毒載量組與健康對(duì)照組間血清PI4KA的含量差異有顯著統(tǒng)計(jì)學(xué)意義(F=119.958,P0.01),并且不同病毒載量組都顯著高于對(duì)照組。隨著HBV DNA載量的升高,PI4KA的含量增加,兩者呈正相關(guān)(r=0.758, P0.01); HepG2.2.15細(xì)胞中PI4KA mRNA的表達(dá)水平較HepG2高,差異有統(tǒng)計(jì)學(xué)意義(P0.05);在細(xì)胞培養(yǎng)上清液中,HepG2和HepG2.2.15細(xì)胞PI4KA的含量分別為1.07±0.54ng/ml和1.63±0.43ng/ml, HepG2.2.15細(xì)胞中PI4KA的含量高于HepG2細(xì)胞(-2.29,P0.05)。 結(jié)論:慢性乙型肝炎患者血清PI4KA的含量與HBV DNA載量密切相關(guān);HBV能夠在mRNA和蛋白水平上調(diào)肝癌細(xì)胞PI4KA的表達(dá);在臨床水平和細(xì)胞水平證實(shí)PI4KA與乙型肝炎病毒復(fù)制密切相關(guān),乙型肝炎病毒可能通過(guò)上調(diào)宿主因子PI4KA的表達(dá)來(lái)促進(jìn)自身病毒的復(fù)制。
[Abstract]:Objective: to investigate the relationship between PI4KA and viral load in patients with chronic hepatitis B, to observe the difference of PI4KA expression in HepG2 and HepG2.2.15, and to explore the effect of HBV on PI4KA expression.
