云南臨滄HIV陽性者血液中弓形蟲SAGSAG2基因位點分析

【摘要】:目的初步了解云南臨滄HIV陽性者血液中弓形蟲的基因型特征。方法運用巢式PCR法對弓形蟲SAG2基因的兩個片段分別進行基因提取、擴增,然后對PCR產(chǎn)物進行回收純化、酶切及測序,并與弓形蟲基因I型標(biāo)準(zhǔn)株進行對比鑒定。結(jié)果從170份HIV陽性者全血樣本中分別擴增出35個弓形蟲SAG2基因(241、221 bp),從其中擴增陽性的全血樣本中各選擇4份分別進行酶切,擴增出目的基因片段,進一步從酶切樣本中選擇2份與弓形蟲基因標(biāo)準(zhǔn)株RH株進行對比,酶切SAG2基因(241 bp)顯示血液樣本基因型為Ⅰ型或Ⅱ型,酶切SAG2基因(221 bp)確定基因型均為Ⅰ型。結(jié)論初步確定云南臨滄HIV陽性者弓形蟲基因型以Ⅰ型為主。
[Abstract]:Objective to investigate the genotypic characteristics of Toxoplasma gondii in the blood of HIV positive patients in Lincang, Yunnan Province. Methods two fragments of SAG2 gene of Toxoplasma gondii were extracted and amplified by nested PCR method. The PCR products were purified, digested and sequenced, and compared with the standard strain of Toxoplasma gondii gene type I. Results 35 samples of Toxoplasma gondii SAG2 gene were amplified from 170 whole blood samples with positive HIV. Four samples of Toxoplasma gondii SAG2 gene were digested by enzyme digestion, and the target gene fragments were amplified. Furthermore, two strains of Toxoplasma gondii gene standard strain RH were selected from the digested samples. The results showed that the genotypes of blood samples were type 鈪,
