肝組織中NLRP3炎癥小體活化與日本血吸蟲感染小鼠肝纖維化程度的相關(guān)性

【摘要】:目的探討B(tài)ALB/c小鼠肝組織中NOD樣受體蛋白3(NOD-like receptor protein 3,NLRP3)炎癥小體活化與日本血吸蟲(Schistosoma japonicum)感染所致肝纖維化程度的關(guān)系。方法 6~8周齡BALB/c雌性小鼠48只,分為未感染組(n=8)、感染后第5、6、8和12周組(n=10)。感染組每鼠經(jīng)腹部皮膚貼片法感染日本血吸蟲尾蚴(20±3)條。取小鼠肝臟組織,用天狼星紅染色法檢測肝組織切片中膠原的含量。抽提肝組織RNA,逆轉(zhuǎn)錄成cDNA,實(shí)時(shí)熒光定量PCR(qPCR)檢測NLRP3、凋亡相關(guān)微粒樣蛋白(apoptosis associated speck-like protein containing a CARD domain,ASC)、半胱天冬酶原1(pro-caspase-1)、白細(xì)胞介素-1β前體(proIL-1β)和IL-18前體(pro-IL-18)的RNA表達(dá)水平。用免疫組化染色結(jié)合H-score評分法檢驗(yàn)小鼠肝臟半胱天冬酶1(caspase-1)p10和IL-1β的蛋白質(zhì)表達(dá)水平。結(jié)果天狼星紅染色結(jié)果顯示,日本血吸蟲感染后第5、6、8、12周小鼠肝臟切片的膠原面積百分比在第8周最高(29.66±1.07)%,其后依次為第6周(21.69±1.24)%、第12周(11.98±0.95)%和第5周(1.76±0.34)%。qPCR結(jié)果顯示,與未感染組相比,感染后第5、6、8、12周小鼠肝臟NLPR3、ASC、pro-caspase-1、pro-IL-1β以及pro-IL-18的相對表達(dá)量為:NLRP3分別為8.12±0.66(P0.01)、24.13±2.81(P0.01)、14.86±0.35(P0.01)、6.74±0.67(P0.01),ASC為0.82±0.14、7.12±0.90、3.08±0.87、4.13±0.93(P0.01),pro-caspase-1為1.14±0.72、2.53±0.46(P0.05)、3.16±0.80(P0.01)、2.19±0.87,pro-IL-1β為9.95±1.04、117.76±10.01(P0.01)、36.98±11.73(P0.01)、7.74±2.27,pro-IL-18為2.42±0.36、1.85±0.11、1.74±0.10、1.69±0.15。免疫組化結(jié)合H-score評分的結(jié)果顯示,感染后第5、6、8和12周小鼠肝臟的caspase-1 p10和IL-1β的H-score值分別為2.80±0.12、2.10±0.06、8.57±1.00(P0.01)、4.55±0.50(P0.01)和0.13±0.13、0.20±0.00、3.30±0.59(P0.01)、1.80±0.43。結(jié)論日本血吸蟲感染小鼠的肝纖維化程度在第8周最明顯,繼而是第6周、第12周和第5周;NLRP3炎癥小體在感染第6周、第8周顯著活化;NLRP3炎癥小體的活化與日本血吸蟲感染所致的肝纖維化程度呈正相關(guān)。
[Abstract]:Objective to investigate the relationship between the activation of inflammatory corpuscles of NOD like receptor protein 3(NOD-like receptor protein 3 (NLRP3) and the degree of hepatic fibrosis induced by Schistosoma japonicum (Schistosoma japonicum) infection in BALB/c mice.Methods Forty-eight female BALB/c mice aged 6 to 8 weeks were divided into two groups: the uninfected group (n = 8), the 5th week group (n = 6) and the 12th week group (n = 10).In the infected group, 20 鹵3 Schistosoma japonicum cercariae were infected through abdominal skin patch method.The contents of collagen in liver sections of mice were detected by Sirius red staining.Immunohistochemical staining and H-score score were used to detect the expression of 1(caspase-1)p10 and IL-1 尾 in liver of mice.Results the results of Sirius red staining showed that the percentage of collagen area in liver slices of mice at the 5th week after schistosomiasis infection was the highest at the 8th week (29.66 鹵1.07), followed by the sixth week (21.69 鹵1.24), the 12th week (11.98 鹵0.95%) and the fifth week (1.76 鹵0.34)%.qPCR).鎰熸煋鍚庣5,6,8,12鍛ㄥ皬榧犺倽鑴廚LPR3,ASC,pro-caspase-1,pro-IL-1尾浠ュ強(qiáng)pro-IL-18鐨勭浉瀵硅〃杈鵑噺涓,
