日本血吸蟲感染鼠肝組織與腸黏膜二者病變發(fā)生發(fā)展的相關(guān)性研究

【摘要】:建立日本血吸蟲感染小鼠不同病期模型,探討肝組織和腸黏膜二者間在病變發(fā)生發(fā)展上的相關(guān)性,以期為血吸蟲病情嚴(yán)重程度和預(yù)后判斷提供實(shí)驗(yàn)依據(jù)及評估方法。研究方法:購自實(shí)驗(yàn)動物公司的健康昆明雄性小鼠50只,隨機(jī)取10只鼠作健康對照組,余40只鼠均按常規(guī)法經(jīng)腹部皮膚感染日本血吸蟲尾蚴(20尾/鼠);感染后按常規(guī)分籠喂養(yǎng)。除感染后第12周隨機(jī)取10只小鼠用吡喹酮(300mg/kg.d)一次灌藥作殺蟲治療外,其余小鼠分別在感染后第6周,第12周和第21周各隨機(jī)取10只小鼠和治療后第9周小鼠作觀察。所有鼠按設(shè)計時點(diǎn):收集血清用ELISA法作TGF-β1和ATP酶的濃度測定;解剖小鼠,仔細(xì)查檢肝門靜脈及腸系膜靜脈內(nèi)血吸蟲蟲體,并記錄每鼠蟲體數(shù),取出肝臟和全結(jié)腸標(biāo)本做大體標(biāo)本觀察記錄后,均作石蠟包埋切片及HE染色,比較觀察和分析其組織病理學(xué)變化。結(jié)果顯示:不同病期血吸蟲病小鼠模型,感染成功率為100%;其病變在感染后6w組急性蟲卵肉芽腫形成,12周出現(xiàn)肝纖維化,21周出現(xiàn)肝硬變,各病期的肝和腸二者病變存在相關(guān)性,取病理學(xué)分級評分指標(biāo)經(jīng)Spearman秩相關(guān)分析的結(jié)果呈正相關(guān)(rs=0.892,P0.01)。對照組、6w組、12w組、21w組和治療組的小鼠血清中ATP酶濃度分別為207.74±4.98nmol/L、302.96±5.54nmol/L、501.72±7.12nmol/L、505.01±5.85nmol/L、211.85±5.51nmol/L。除對照組與治療組、12w組與21w組的差異無統(tǒng)計學(xué)意義外,其余各組間的差異均有統(tǒng)計學(xué)意義(P0.05)。小鼠血清中TGF-β1濃度隨感染周期的延長而上升,用藥后顯著下降,用藥組與對照組間的差異無統(tǒng)計學(xué)意義(P0.05)。研究結(jié)論:日本血吸蟲感染后不同病期小鼠的結(jié)腸組織學(xué)評分值與肝臟病理分級呈正相關(guān);血清中TGF-β1和ATP酶濃度隨病程期延長有增加趨勢,在治療后降低,提示其可能具有病情評估意義。圖8幅,表11個,參考文獻(xiàn)95篇。
[Abstract]:To study the relationship between liver tissue and intestinal mucosa in the pathogenesis of schistosomiasis japonicum infection in mice at different stages. In order to provide experimental basis and evaluation method for the severity of schistosomiasis and its prognosis, 50 healthy Kunming male mice were purchased from experimental animal company and 10 mice were randomly selected as healthy control group. The remaining 40 mice were infected with 20 cercariae of Schistosoma japonicum through abdominal skin according to the routine method, and were fed in normal cage after infection. 10 mice were randomly given praziquantel (300 mg / kg 路d) for insecticidal treatment at 12 weeks after infection. The other mice were randomly selected for observation at the 6th, 12th and 21st weeks after infection and at the 9th week after treatment. All the mice were collected for the determination of TGF- 尾 1 and ATP enzyme by ELISA method, and the mice were dissected. The body of Schistosoma japonicum in portal vein and mesenteric vein of liver was carefully examined, and the number of worm bodies per mouse was recorded. After taking out the liver and whole colon for general observation and recording, all the specimens were stained with HE and paraffin embedded sections. The histopathological changes were observed and analyzed. The results showed that the success rate of infection was 100% in mice with schistosomiasis at different stages, and hepatic cirrhosis occurred at 12 weeks after acute egg granuloma formation and 21 weeks after infection. There was a correlation between liver and intestine lesions in each stage of the disease. The pathological grading index was positively correlated with the results of Spearman rank correlation analysis. The serum ATP enzyme concentration in the control group was 207.74 鹵4.98nmol / L 302.96 鹵5.54nmol / L 501.72 鹵7.12nmol / L 505.01 鹵5.85nmol / L = 211.85 鹵5.51nmol / L, except that there was no significant difference between the control group and the control group (12w) and the 21w group (P < 0.01). The serum TGF- 尾 1 concentration of mice increased with the prolongation of infection cycle, and decreased significantly after treatment. There was no significant difference between the treatment group and the control group (P 0.05). Conclusion: the colonic histological score of mice with different stages of schistosomiasis infection was positively correlated with the pathological grade of liver. The concentrations of TGF- 尾 1 and ATP in serum increased with the prolongation of the course of disease and decreased after treatment, suggesting that TGF- 尾 1 and ATP enzyme concentrations may be of significance for the evaluation of the disease. 8 figs, 11 tables, 95 refs.
